BDNF对脊神经根结扎大鼠脊髓背角星形胶质细胞活化的影响

摘    要:目的观测脊神经根结扎(SNL)大鼠脊髓背角中脑源性神经营养因子(BDNF)与星形胶质细胞活化标记蛋白胶质细胞纤丝酸性蛋白(GFAP)表达的变化,以及BDNF清除剂TrkB/Fc对GFAP表达和疼痛的影响。方法取18只SD大鼠,随机分为空白对照组、假手术组和SNL组(n=6)。分别于术前、术后第1~6天测定大鼠50%机械缩爪阈值(50%PWT),末次测定后取腰膨大处脊髓背角经Western blotting检测GFAP和BDNF的表达。另取18只SD大鼠,随机分为对照组、SNL+安慰剂组和SNL+TrkB/Fc组(n=6),其中对照组仅行鞘内置管术;SNL+安慰剂组和SNL+TrkB/Fc组分别在行SNL术前,予鞘内注入PBS或TrkB/Fc 10μL,术后1次/d×6 d,每次注药前1 h测定50%PWT,末次给药后1 h取大鼠腰膨大处脊髓背角经Western blotting检测GFAP的表达。结果术后第1~6天,SNL组手术同侧后肢50%PWT均较术前基础值显著下降(P<0.01);术后第6天,SNL组手术同侧脊髓背角BDNF和GFAP表达均较空白对照组显著升高(P<0.01);SNL+TrkB/Fc组大鼠手术同侧50%PWT与基础值比较无明显变化(P>0.05),手术同侧脊髓背角GFAP表达与对照组比较差异无统计学意义(P>0.05)。结论 BDNF活化的脊髓星形胶质细胞对SNL诱发的神经病理性疼痛具有调控作用。
