锰对大鼠睾丸毒性效应的研究

摘    要:目的研究氯化锰对大鼠睾丸形态学及精子的毒性效应,探讨锰对大鼠生精功能损伤作用的机理。方法雄性SD大鼠48只,随机分为6组:空白对照组,15mg/kg和30mg/kgMnCl2组,8只/组。15mg/kgMnCl2组和30mg/kgMnCl2组分别染锰(MnCl2·4H2O)4wk和6wk,空白对照组给予等容生理盐水,给锰与生理盐水途径均为腹腔注射,5d/wk,1次,d,大鼠分别于第4wk末和第6wk术处死,取睾丸作以下检测:①观察睾丸形态学改变;②检测睾丸脏器系数;③精子数量。结果①与空白对照组比较,染锰15mg/kg、4wk即可对大鼠生精小管形态结构造成损伤,且随染锰剂量和时间的增加生精小管损伤越严重;染锰4w30mg/kgMnCl2组睾丸脏器系数显著降低(P〈0.01),各染锰组精子数量均显著降低(P〈0.01);②染锰剂量相同,染锰6wk组与染锰4wk组比较,精子数量均显著降低(P〈0.01);③染锰时间相同,30mg/kgMnCl2组与15mg/kgMnCl2组比较,精于数量均显著降低(P〈0.01)。结论染锰15mg/kg、4wk即可对大鼠睾丸形态学和精子生成造成损伤,并存在一定的剂量-效应和时间-效应关系,证实了过量锰对雄性大鼠睾丸具有明冠毒性效应。

Abstract:Objective Tc Study on Toxic effect on the morphology of testis and sperm in rats exposed to manganese Methods The 48 male rats were randomly divided into one control group and two manganese chloride (MnCl2)(15,30mg/ kg) groups. After exposed to manganese for 4 and 6 weeks respectively, the morphological changes of rats testis were observed , the weight coefficient of testis were detected, quantity of sperm was counted. Results compared with control group , damage of tats testis was serious with increase of dose and time exposed to manganese; the weight coefficient of testis was decreased in 4w 30mg/kg MnCl2 group significantly(P〈0.01 ), the quantity of sperm was decreased significantly in every manganese exposure groups (P〈0.01). Among manganese exposure groups o same dose, the quantity of sperm was decreased more siguificandy in 6w manganese exposure groups than in 4w manganese exposure groups (P〈0.01).Among manganese exposure groups of same time, the quantity of sperm was decreased more signficantly in 30mg/kg manganese exposure groups than it in 15 mg/kg manganese exposure groups (P〈0.01).Conclusions ①15mg/kg MnCl2 exposure for 4w The damage of the morphology of testis and quantity of sperm were significant,but dose and time effects were found . ② It was suggested dtat excessive manganese can have serivous toxic effect on testis in rats.
