白细胞介素8在室管膜肿瘤细胞中的表达及分泌的意义

摘    要:目的 探讨室管膜肿瘤中白细胞介素8(IL-8)表达和分泌的意义。方法 采用免疫组织化学法检测50例手术切除的室管膜肿瘤标本中IL-8表达水平;双抗体夹心ELISA法测定原代培养的室管膜肿瘤细胞上清液中IL-8含量。结合患者的一般状况,对不同组织学分型、病理分级及侵袭性的肿瘤的IL-8表达和分泌量进行统计学检验。结果 正常脑组织IL-8表达为阴性或弱阳性,而室管膜肿瘤IL-8表达均为阳性。间变型或恶性室管膜瘤每高倍视野中IL-8阳性细胞数目为417±38个,多于细胞和上皮型室管膜瘤(P<0.01);IL-8阳性染色强度为56±7.5AU,高于细胞和上皮型室管膜瘤(P<0.01)。间变型或恶性室管膜瘤细胞上清液中IL-8的含量为638μg/L±87μg/L,高于细胞和上皮型室管膜瘤(298μg/L±48μg/L,P<0.01)。侵袭性室管膜肿瘤中IL-8的表达和分泌水平高于非侵袭性室管膜肿瘤(P<0.01)。高恶性室管膜肿瘤细胞中IL-8的表达和分泌高于低恶性或良性室管膜瘤(P<0.01)。IL-8的表达和分泌水平与患者性别、年龄等无明显相关性。结论 高恶性室管膜肿瘤中IL-8表达和分泌显著高于低恶性或良性室管膜肿瘤细胞。IL-8的表达和分泌可能与室管膜肿瘤的发生和发展密切相关,可作为判断室管膜肿瘤恶性程度及患者预后的诊断指标之一。

Abstract:Objective To detect the expression and secretion of IL 8 in the ependymal tumor tissue and in primarily cultured cells, and to explore the relationship between IL 8 and ependymal tumor. Methods Expression of IL 8 in 50 ependymal tumor specimens was examined by immunohistochemistry. The ependymal tumor tissue obtained during operation was primarily cultured and IL 8 secreted by cultured cells were examined by clouble antibody 5 and wich ELISA. The relations of expression of IL 8 to ependymal tumors of different histological degree, and invasiveness was analyzed statistically. Results All the ependymal tumor tissue expressed IL 8, but the number of IL 8 positive cells in the anaplastic and malignant ependymoma was evidently larger than that in ependymoma ( P <0 01). All the cells cultured from the ependymal tumor secreted IL 8, but the liquid supernatant of anaplastic and malignant ependymoma cells contained nore IL 8 than that of ependymoma cells ( P <0.01). The expression and secretion of IL 8 had no close relati on to age and sex of patient. Conclusion The expression and secretion of IL 8 may be involved in the progression of ependymal tumor, so IL 8 can be used as an indicator to detect the malignant degree of ependymal tumor.
