摘    要:富颈癌 DNA 在两种特异引物和 DNA 聚合酶的存在下,经35轮循环的变性、退火和链的延伸反应,c-Ha-ras 癌基因100bp 片段被放大约10~6倍。反应产物经琼脂糖或聚丙烯酰胺凝胶电泳可见清晰的扩增带。

Abstract:This paper described the method with which that 100 bp fragment of the first exon of c-Ha-ras is amplified by the Polymerase chain reaction. The DNA of cervical cancer was denatured, then the primers annealed to the target sequence, and DNA polymerase catalyzed the extention of the primers. After thirty-five cycles, the speific fragment was amplified 10~6 times. Analyzed by agarose gel elec- trophoresis or acrylamide gel electrophoresis, the band of DNA was visible under the UV light. This technique may be used for diagnosis of the c-Ha-ras oncogene and the final reaction mixture may be used for the diagnosis of the c-Ha-ras oneogene and the final reaction mixture maybe used for the anal- ysis of mutation of the c-Ha-ras oncogene.
