染料木黄酮对ICR小鼠抗氧化能力及AGE水平的影响

摘    要:目的 了解染料木黄酮 (Genistein)对 ICR小鼠组织抗氧化能力及高级糖基化终末产物 (AGE)水平的影响。方法 分别用不同剂量的染料木黄酮灌胃 1 6月龄的 ICR小鼠 30 d,然后分别对小鼠脑、肝等组织 AGEs、总活性氧 (ROS)、丙二醛 (MDA)水平以及谷胱甘肽过氧化物酶(GSH- Px)活性进行测定。结果 染料木黄酮灌胃小鼠肝组织 AGEs、ROS及 MDA水平均显著下降 (P<0 .0 5) ,GSH- Px活性升高 (P<0 .0 5) ;灌胃组小鼠脑组织 AGEs水平及 GSH- Px活性均与对照组小鼠无显著差异 ,而 ROS和 MDA水平不同程度下降 (P<0 .0 5)。结论 染料木黄酮可提高小鼠的抗氧化能力及肝组织细胞中 GSH- Px活性 ,从而有效降低肝组织中的氧化应激水平 ,抑制肝组织中 AGEs的形成 ;而其对脑组织 AGEs水平及GSH- Px活性均未产生明显影响。

Abstract:Objective To investigate the effects of genistein on the antioxygenic propertys and the levels of advanced glycation end products (AGEs) in the tissues of ICR mice Methods The levels of AGEs, reactive oxygen species (ROS) and maleic dialdehyde (MDA), the activity of glutathione peroxidase (GSH Px) of the tissue of ICR mice administrated with different doses of genistein were measured Results The levels of AGEs, ROS and MDA of the liver tissue of the treated mice decreased significantly ( P <0 05), and the activity of GSH Px of the liver tissues increased There is no difference of AGEs and GSH Px in the brain tissue between the treated mice and the control mice, but the levels of ROS and MDA of the brain tissue of the treated mice decreased compared with the control mice Conclusions Genistein may improve the antioxygenic property of ICR mice and promote the activities of GSH PX of the liver tissue, which can be beneficial for removing free radicals (FR) and ROS and blocking formation of AGEs However, there are no significant effects of genistein on the AGEs level and GSH PX activity in the brain tissue of ICR mice
