摘 要: | 目的 研究连梅汤(Lianmei Decoction,LMT)对2型糖尿病(T2DM)的治疗作用。方法 将C57BL/6J设为空白组、db/db糖尿病小鼠分为对照组,二甲双胍组,连梅汤低、中和高剂量组。收集不同组别小鼠的血清进行肝功能相关指标TC、AST、ALT、HDL-c、LDL-c和TG检测,验证连梅汤对小鼠血糖和血脂的调节作用,最后对实验小鼠粪便进行16SrDNA测序。结果 db/db小鼠与C57BL/6J比较血糖和血脂显著升高,研究模型出现T2DM。实验结果表明,在连梅汤的治疗下,与C57BL/6J小鼠比较,连梅汤对血糖、血清中FFA、血清中INS均有一定的抑制作用。db/db糖尿病小鼠TC、AST、ALT、HDL-c、LDL-c和TG在治疗组都有趋于正常的趋势,且在低剂量治疗组的治疗效果优于阳性药物二甲双胍。HE染色显示连梅汤不同剂量治疗组结肠未见明显病理改变。肠道菌群结果显示,连梅汤能够显著提高拟杆菌和降低厚壁菌门的丰度。虽然连梅汤在属水平降低了肠道微生物的相对丰度,但是却扩大了肠道微生物的总群多样性。结论 该研究证明了连梅汤可能通过影响肠道菌群对db/db糖尿病小鼠的肝脏功能和血糖血脂具有一定的调节作用,为连梅汤的临床治疗提供了一定的理论依据。

Effects of Lianmei Decoction (LMT) on Blood Glucose and Intestinal Flora in Mice with Diabetes Mellitus |

Abstract: | Objective To study the therapeutic effect of LMT on type 2 diabetes (T2DM).Methods C57BL/6J and db/db diabetic mice were selected as the study subjects, and C57BL/6J was set as the blank group, db/db diabetic mice were divided into control group, positive drug group, low, medium, and high-dose group. The serum of mice in different groups was collected for liver function related indexes TC, AST, ALT, HDL-c, LDL-c, and TG to verify the regulating effect of LMT on blood glucose and blood lipid in mice. Finally, 16SrDNA sequencing was performed on the feces of experimental mice to analyze the changes of intestinal flora and to explore whether it affected the changes of blood glucose in db/db mice through the liver.Results Compared with C57BL/6J, db/db mice had significantly higher blood sugar and blood lipids, abnormal liver function-related indicators, and T2DM in the research model. The experimental results showed that under the treatment of Lianmei Decoction, compared with C57BL/6J mice, Lianmei Decoction had a certain inhibitory effect on blood sugar, FFA in serum, and INS in serum. The TC, AST, ALT, HDL-c, LDL-c, and TG of these db/db diabetic mice tended to be normal in the treatment group, and the treatment effect in the low-dose treatment group was better than that of the positive drug metformin. HE staining showed that there were no obvious pathological changes in the colon in different doses of LMT. The results of intestinal flora showed that LMT was able to significantly increase the abundance of Bacteroidetes and decrease the abundance of Firmicutesin phylum. Although the relative abundance of intestinal microorganisms was reduced at the genus level by LMT, the total population diversity of intestinal microorganisms was expanded.Conclusion This study demonstrated that LMT could regulate liver function and blood glucose and lipids in db/db diabetic mice by affecting intestinal flora, which provides some theoretical basis for the clinical treatment of LMT. |
