补肾益气活血中药对大鼠脑缺血再灌注损伤后VEGF的动态变化与BBB通透性改变的影响

摘    要:目的探讨补肾益气活血中药对大鼠脑缺血再灌注损伤后血管内皮生长因子(VEGF)的动态变化与血脑屏障(BBB)通透性改变的影响。方法将大鼠随机分为假手术组、生理盐水组、中药组,采用线拴法建立脑缺血模型。采用伊文思蓝(EB)荧光测定法分别对大鼠脑组织进行缺血再灌注1、3、7d后的EB含量变化情况进行检测;采用免疫组织化学方法测定大鼠脑缺血再灌注不同时间点VEGF的表达情况。结果生理盐水组和中药组大鼠再灌注1、3、7d后湿脑组织中EB含量分别为(4.69±0.97)μg/g、(5.89±1.13)μg/g、(3.97±0.67)μg/g和(3.32±1.43)μg/g、(4.27±1.48)μg/g、(3.19±0.64)μg/g,均高于假手术组的(2.12±0.45)μg/g(P〈0.05)。中药组大鼠湿脑组织中EB含量在同一时间点均明显低于生理盐水组(P〈O.05)。结论补肾益气活血中药可通过对血脑屏障功能的改善减轻血管源性脑水肿。

Abstract:Objective To investigate the effect of bushenyiqihuoxue traditional Chinese medicine on the expressiorl of VEGF protein after reperfusiun of transiently cerebral ischemia and the transportation of blood-brain barrier in rat. Methods All rats tethered cord ischemia model were randomly divided into sham group,saline group,traditional Chinese medicine group. The level of EB ischemia-reperfusion cerebral were detected by Evans Blue (EB) fluorescence assay after 1,3,7days. The expression of VEGF of cerebral ischemia and reperfusion in rats were observed at different time. Results The level of EB of the saline group, traditional Chinese medicine group after reperfusiun lday, 3day,7day were (4.69±0.97) μg/g, (5.89±1.13) μg/g, (3.97±0.67)μg/g and (3.32±1.43)μg/g, (4.27±1.48)μg/g, (3.19±0.64) μg/g those were higher than those of sham group(P 〈0.05).The level of EB of Chinese medicine group was lower than of the saline group (P 〈0.05). Conclusion Bushenyiqihuoxue traditional Chinese medicine decreases vasoactive cerebral edema by improving the function of BBB.
