温郁金中二萜类化合物C抑制结肠腺癌细胞增殖并诱导凋亡的实验研究

摘    要:背景:温郁金是临床上常用的中药,其抗肿瘤作用正日益受到关注。目的:研究温郁金中二萜类化合物C对人结肠腺癌SW620细胞的抑制作用及其机制。方法:将不同浓度(10-70μg/m1)二萜类化合物C作用SW620细胞24、48和72h,并以5.氟尿嘧啶(5.Fu)作为阳性对照。采用M1Tr法检测细胞增殖情况.倒置相差显微镜下观察细胞形态学变化:以流式细胞术检测浓度分别为0、40、55和70μg/ml二萜类化合物C作用24h和48h后对SW620细胞周期和凋亡率的影响。结果:二萜类化合物C能以时间-浓度依赖性方式抑制SW620细胞增殖.其抑制能力均显著高于相同时间点和浓度5-Fu组.差异有统计学意义(P〈0.01);倒置相差显微镜下发现二萜类化合物C作用后的SW620细胞呈典型的凋亡改变。流式细胞术显示二萜类化合物C能阻滞SW620细胞于G,和S期,减少G,期比例,并诱导细胞凋亡。结论:温郁金中二萜类化合物C对人结肠腺癌SW620细胞的增殖有显著抑制作用,并可阻滞细胞周期和诱导细胞凋亡.可能是其发挥抗肿瘤作用的机制之一。

Abstract:Background: Radix Curcumae is a commonly used traditional Chinese medicine in clinics,and its antitumor effects have increasingly attracted attention from scientists.Aims: To investigate the inhibitory effects of diterpenoid C extracted from Radix Curcumae on human colonic adenocarcinoma cell line SW620 and its underlying mechanism.Methods: SW620 cells were treated with different concentrations of diterpenoid C(10-70 μg/ml) for 24,48 and 72 hours,respectively,and 5-fluorouracil(5-Fu) served as positive control.Cell proliferation was assessed by MTT assay,and cell morphology was observed under inverted phase contrast microscope.Cell cycle and apoptosis were determined by flow cytometry after SW620 cells were treated with different concentrations of diterpenoid C(0,40,55 and 70 μg/ml) for 24 and 48 hours,respectively.Results: Diterpenoid C inhibited the cell proliferation in time-and dose-dependent manners,its inhibitory effects were significantly higher than 5-Fu at the same time point and concentration(P<0.01),and typical apoptotic changes could be seen under inverted phase contrast microscope.Flow cytometry showed that diterpenoid C arrested SW620 cells at cell cycle G1 and S phases,reduced the proportion at G2 phase,and induced apoptosis.Conclusions: Diterpenoid C extracted from Radix Curcumae significantly inhibits proliferation of human colonic adenocarcinoma SW620 cells,induces cell cycle arrest and apoptosis,which may be one of the underlying mechanisms of its antitumor effects.
