核因子κB在大鼠双侧后肢缺血再灌注后肾损伤中的表达及意义

摘    要:目的探讨核因子κB(NF-κB)在大鼠双侧后肢缺血再灌注(I/R)所致肾损伤中的表达及意义.方法建立大鼠双侧后肢I/R模型;48只大鼠随机分为假手术组(Ⅰ组,n=8)、缺血组(Ⅱ组,n=8)、缺血再灌注组(Ⅲ组,n=32).Ⅱ组在缺血4 h末,Ⅲ组分别于再灌注4,12,24和48 h切取双肾,应用半定量RT-PCR和Westem bolt方法检测肾组织NF-κB p65/I κBβ的mRNA和蛋白产物表达.结果Ⅲ组时各时点NF-κ B p65 mRNA表达较Ⅰ、Ⅱ组增加,在灌注后12 h达高峰.P65蛋白产物表达亦逐渐增加,12 h高峰,可持续到术后48 h.I κ B β的mRNA和蛋白表达在I/R后同样开始增加,24 h的表达量最多.结论NF-κ B/I κ B系统的激活可能在骨骼肌I/R所致肾损伤中发挥重要作用.

Abstract:Objective: To investigate the activation and expression of nuclear factor -κB(NF-κB)/I-κB system in renal injury induced by ischemia/reperfusion (I/R) of skeletal muscle in rats. Methods: The rat model of hind limbs I/R injury was established and 48 rats were divided into three groups: sham operation group (group Ⅰ, n =8), ischemia group(group Ⅱ, n =8), I/R injury group (group Ⅲ, n =32). I/R group were divided by 4 hours, 12 hours, 24 hours and 48 hours after reperfusion. The changes of mRNA and protein expression of NF-κB p65 / IκBβ in kidney were studied by RT-PCR or Western blot. Results: In group Ⅲ, NF-κB p65 mRNA rose quickly after reperfusion and reached the peak 12 hours after reperfusion (36.5%+6.6%), and p65 detected by Western blot reached the peak 12 hours after reperfusion (66.0%+10.3%) too and then decreased slowly, still high level 48 hours after reperfusion (21.0%+5.4%). Expression of IκBβ also increased early reperfusion and mostly on 24 hours after reperfusion. Conclusions: NF-κB / IκBβ system may be an important role in the damage of renal function induced by skeletal muscle I/R injury.
