摘    要:目的探讨高效液相色谱法(HPLC)测定盐酸法舒地尔含量的可行性。方法采用高效液相色谱法测定自制盐酸法舒地尔含量。色谱柱:phecda C18(250 mm×4.6 mm,5μm),以1.0%三乙胺水溶液(用磷酸调节pH值至7.0)-甲醇(50∶50)为流动相;检测波长为275 nm,流速:1.0 ml/min;进样量:20μl;柱温:25℃。结果 HPLC法测定盐酸法舒地尔的浓度在0.048 4~0.242 0 g/L的范围内,浓度与峰面积有良好的线性关系(r=0.9994,n=5),平均回收率为(99.4±0.47)%,RSD为0.47%(n=9)。结论 HPLC法测定简单,专属性强、灵敏度高,可以用于盐酸法舒地尔的含量测定。

Abstract:Objective To investigate the feasibility of determination of fasudil hydrochloride content by high performance liquid chromatography( HPLC). Methods HPLC was used to determine the content of a self-prepared fasudil hydrochloride solution.Separation was performed on the phecda C18 column( 250 mm × 4. 6 mm,5 μm) with the mobile phase of triethylamine solution( the pH value adjusted to 7. 0 using phosphoric acid)- methanol( 50∶ 50),at a detection wave length of 275 nm,a flow rate of 1. 0 ml /min,feed volume of 20 μl and column temperature of 25 ℃. Results With HPLC,the content level was within 0. 0484- 0. 2420 g /L.Linear relationship could clearly be noted between the content and the peak area( r = 0. 9994,n = 5). Average recovery rate was( 99. 4 ±0. 47) % and relative standard deviation( RSD) was 0. 47%( n = 9). Conclusion HPLC was a simple,sensitive and specific detection method,which could be used for the determination of fasudil hydrochloride content.
