四君子汤(生晒参/红参)对H2O2诱导H9c2心肌细胞凋亡的保护作用研究

摘    要:该研究首先筛选了四君子汤(红参)等药液组体外干预H9c2心肌细胞的给药浓度,优选其中高、中、低3个剂量组进行后续实验;建立了体外H2O2诱导H9c2心肌细胞凋亡模型以考察四君子汤(生晒参/红参)对其保护作用,从而为优选用于临床治疗缺血性心脏病的四君子汤中的人参炮制品提供参考,以更好地体现其疗效;并考察其对SOD,MAD和LDH等指标影响以初步阐明作用机制。结果表明,四君子汤中用红参较生晒参对H2O2诱导心肌细胞损伤保护作用为好,二者均可提高SOD活性,减少MDA产生和LDH释放,从而明显减少心肌细胞凋亡数量,起到一定的保护作用。

Abstract:In this study, efforts were made to screen out the drug concentration of Sijunzi decoction (red ginseng) for in vitro intervention of H9c2 cardiomyocytes, select high, medium and low groups for subsequent experiments, establish the H2O2-induced myocardial cell apoptosis to investigate the protective effect of Sijunzi decoction (white/red ginseng), provide reference ginseng ingredients in Sijunzi decoction used to treat ischemic heart disease and reflect its curative effect, and observe its impacts on SOD, MAD, LDH and other indexes to preliminarily define the action mechanism. According to the results, red ginseng in Sijunzi decoction showed a better protective effect on H2O2-induced myocardial cell injury than that of white ginseng. Both of them could enhance SOD activity and reduce MDA production and LDH release, so as to significantly reduce the amount of apoptotic myocardial cells and play protective role.
