| 摘 要： | 目的：探讨碱性成纤维细胞生长因子（basic fibroblast growth factor，bFGF）反义寡核苷酸对肺癌细胞凋亡的影响。方法：以脂质体作为转染载体，转染bFGF反义寡核苷酸于SH-77细胞，用Western blot方法检测bFGF蛋白的表达，用流式细胞仪检测SH-77细胞凋亡率。结果：与随机对照序列组相比,bFGF反义寡核苷酸对SH-77细胞中bFGF蛋白表达有明显抑制作用，P〈0．05；可使SH-77细胞凋亡率明显增加，P％0．01。结论：bFGF在肺癌细胞凋亡中有重要调节作用，可作为肺癌增敏治疗的有效靶点。

| Abstract: | OBJECTIVE:To investigate the cell cycle chang of lung cancer cells and the role of antisense oligonucleotide targeting bFGF.METHODS:The inhibition of bFGF protein expression was investigated by Western blot in best condition of transfection of antisense oligonucleotide targeting bFGF in SH-77 cells.Apoptosis rate of SH-77 cells was investigated by flow cytometer.RESULTS:The treatment with antisense oligonucleotide of bFGF not only reduced the expression of bFGF(P<0.05),but also increased the apoptosis of SH-77 cells,P<0.01.CONCLUSION:bFGF may take part in the apoptosis regulation of lung cancer cells and be used as a target of lung cancer therapy. |
