摘 要: | 目的:观察痰热清对Wistar大鼠急性放射性肺炎肺组织病理学改变及血清转化生长因子-β1(TGF-β1)水平的变化。方法:45只雌性Wistar大鼠随机分为空白组、模型组、痰热清治疗组(治疗组),每组动物均为15只。用6MV-X线直线加速器对大鼠双肺进行单次照射25Gy。分别于照射后2、4、6周三个时间点,每组各随机取5只大鼠活体观察和取材。分别取1mL和4mL腹主动脉血于试管中用于检测血常规和TGF-β1;测定肺泡灌洗液(BALF)中性粒细胞计数;分离大鼠肺脏经宏观检查并称重后,计算肺系数;取右上肺组织切片,行HE染色进行光镜观察病理改变。结果:模型组大鼠于照射后4周出现较严重的放射性皮肤损害,治疗组大鼠皮损相对较轻。模型组大鼠肺组织于照射后2周就出现急性放射性肺炎改变,第6周时肺间质增厚明显,肺泡腔纤维素渗出。除空白组外,治疗组大鼠肺组织放射性肺炎反应最轻,而且肺系数和血常规及BALF中白细胞数相对较低(P<0.05),血清TGF-β1较模型组低(P<0.05)。结论:痰热清可抑制TGF-β1表达,减轻急性放射性肺炎的炎症反应,痰热清对急性放射性肺炎具有较好的治疗作用。

Experimental Study on the Treatment of Acute Radiation Pneumonia of Wiatar Rats by Tanreqing Injection |

Abstract: | Objective:To observe Tanreqing in treating acute radiation pneumonia of rats and to investigate the changes of TGF-β1 of serum and pathological changes of lung tissue.Methods:Forty five Wistar female rats were randomly allocated into blank control group, model group, Tanreqing treatment group. Each group had 15 rats, The lungs of all rats except those in the blank group were irradiated by 6MV-X ray linear accelerator for 25Gy dose.5 rats in the three groups were randomly got to be observed and drawn the specimen at each of the 3 time points(2, 4 and 6 weeks after irradiation). Their arterial blood was obtained to detect blood routine, TGF-β1 .Their BALF was detected to classify the neutrophil.The lungs of rats' were harvested out, calculated the lung coefficient, then sliced and dyed with HE stain to observe the histological changes.Results:The rats in model group had severe irradiation injury of skin 4 weeks after radiation, however, the skin lesion of rats in treatment group was moderately slight. The lung tissue of rats in model group had radiation pneumonia changes 2 weeks after irradiation, the lung stroma was obviously thickening and much cellulose effused in pulmonary alveoli.Except for the blank group, radiation pneumonia of rats in treatment group was slightest, the lung coefficient and the number of leukocyte of blood routine and BALF was lower than control group (P 〈0.05), TGF-β1 was lower than model group (P 〈0.05).Conclusions:Tanreqing can inhibit expression of TGF-β1 to lessen the inflammatory reaction of radiation pneumonia, it shows therapeutic action on acute radiation lung injury. |
