蒙药连翘-4味汤散对CCL4引起急性肝损伤保肝降酶作用有效部位筛选研究

摘    要:目的:研究蒙药连翘-4味汤散对CCL4引起的急性肝损伤保肝降酶作用有效部位筛选.方法:采用0.25% CCL4腹腔注射(标准为0.2mL/20g)方法制备小鼠急性肝损伤模型.造模18h后分别检测正常组、模型组、各提取物组(即石油醚、氯仿、乙酸乙酯、无水乙醇、75%乙醇、水)ig给药,连续7天给药后血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、胆碱酯酶(CHE)、单胺氧化酶(MAO)的含量和活性.结果:与正常组比较,模型组血清AST、ALT、MAO含量明显增高,P<0.01和P<0.05;CHE含量明显降低(P<0.05).各提取物组与模型组比较:石油醚提取组对AST有明显降低作用(P<0.01);氯仿提取组对AST有明显降低作用(P<0.01)、对CHE有明显增高作用(P<0.05);乙酸乙酯提取组对CHE有明显增高作用(P<0.05);无水乙醇提取组对CHE有明显增高(P<0.01);75%乙醇提取组对AST有明显的降低作用(P<0.05)、对MAO明显降低(P<0.01);水提取组对AST、ALT有明显降低作用(P<0.05、P<0.05)、对CHE有明显增高作用(P<0.01)、对MAO有显著降低(P<0.05).结论:综合考虑蒙药连翘-4味汤散水提取物对CCL4引起急性肝损伤具有明显的保肝作用,且本次试验测定的各项指标与模型组比较均有显著差异.

Studies of the Choice of the Effective Part of MongoLian Medicine Lianqiao-4 Ingredient Soup Powder Which Has Effect of Liver Protection and Enzyme Decrease on the Acute Liver Injury Caused by CCl4

Abstract:Objective :To research the choice of the effective part of Mongolian medicine lianqiao - 4 ingredient soup powder which has function of liver protection and enzyme decrease to the acute liver injury caused by ccL/4. Methods:Using intraperitoneal injection of 0.25% CCl4(standard is 0.2mL/20g) to prepare the rat modeLs of liver injury. After making models 18h, detecting the dose of normal group,model group,every extractive group ( aether petrolei, chloroform, acetic ether, absolute ethyl alcohol,75 % alcohol and water), after 7 days' dose, the contents and activities of AST, ALT, CHE, MAO in the blood serum. Result : Compared with the normal group, the contents of blood serum,AST,ALT and MAO in the model group all has a significant increase are P 〈 0.01 and P 〈 0.05 ; the content of CHE has a significant decrease ( P 〈 0.05 ). The extractive groups compared with the model groups:the aether petrolei extractive group has a significant decrease function to the AST( P 〈 0.01 ) ;the chloroform extractive group has an obvious decrease function on AST( P 〈 0.01 ),an obvious increase function on CHE( P 〈0.05) ;acetic ether extractive group has an obvious significant increase function to CHE( P 〈0. 05) ;absolute ethyl alcohol extractive group it has a significant increase on CHE( P 〈0.01 ) ;75% alcohol extractive group has an obvious decrease on AST( P 〈 0.05 ),obvious decrease on MAO ( P 〈 0.01 ) ;water ex- tractive group has an obvious decrease on AST,ALT( P 〈0.05, P 〈0.05),an significant increase on CHE( P 〈0.01 )and an significant decrease on MAO( P 〈 0.05 ). Conclusion: Overall consideration, water extractive group of Mongolian medicine lianqiao - 4 ingredient soup powder has an obvious liver protecting function on the acute liver injury which caused by CCl4. The indexes of this trial all have significant difference compared with model group.
