赞丹-3汤滴丸对大鼠局灶性脑缺血的保护作用及其机制初探

摘    要:目的:研究给予赞丹-3汤滴丸对预防大鼠局灶性急性脑缺血的保护作用.方法:将72只大鼠随机分组:假手术组、缺血模型组、尼莫地平组(0.024 g.kg^-1)、赞丹-3汤滴丸(0.300g.kg^-1,0.150g.kg^-1,0.075g.kg^-1)剂量组,每组12只,持续给药25d后制备大鼠大脑中动脉阻塞缺血模型(MACO),待大鼠清醒后观察大鼠的神经行为学评分,缺血24h后测定血清丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,病理切片观察神经元的形态结构的改变,免疫组织化学方法检测NF-κB蛋白活性的表达.结果:赞丹-3汤滴丸能明显减少局灶性脑缺血大鼠的神经功能评分(P<0.05),降低局灶性脑缺血后血清中MDA含量,提高SOD活性(P<0.01或P<0.05),明显减轻神经细胞形态的病理损伤,并且抑制NF-κB活性的表达(P<0.05).结论:预防给予赞丹-3汤滴丸对大鼠局灶性脑缺血有明显的保护作用,其机制可能是通过抗氧化应激作用,抑制炎症与免疫反应来实现.

Protective Effects of ZanDan-3Tang Dropping Pills on Focal Cerebral Ischemia Injury in Rats and its Mechanism

Abstract:AIM Prevention research grant ZanDan -3Tang Dropping Pill against focal cerebral ischemia. Meth- ods The 72 rats were randomly divided: sham operation group, model group, nimodipine group (0. 024 g. kg^-1 ) , ZanDan - 3 soup pills (0. 300g. kg-1, 0. 150g. kg^-1, 0. 075 g. kg^-1 ) dose groups of 12, 25 days after the last administration, preparation of middle cerebral artery occlusion ischemia model (MACO). After the rats were observed to be awake rat neural behavioral score, isehemia 24 hours after serum malondialdehy deeontent, superoxide dismutase activity, neuronal pathology observed morphological changes in the structure, immunohisto- chemistry was used to detect the activity of NF - KB protein expression.. Results ZanDan - 3 Tang pills can sig- nificantly reduce focal cerebral ischemia in rats neurological score ( P 〈 0.05 ), lower serum after focal cerebral isehemia MDA content, SOD activity increased ( P 〈 0.01 or P 〈 0.05 ), significantly reducing pathological damage nerve cell morphology, and inhibiting the expression of NF - KB activity ( P 〈 O. 05 ). Conclusion Zan- Dan -3Tang Droppin Pills prevention given on cerebral ischemia have a significant protective effect, the mechanism may be through the role of oxidative stress, inhibition of inflammation and immune responses to achieve.
