摘 要: | 目的:探讨藏药湿生扁蕾对SW480细胞凋亡调控因子Bcl-2mRNA及BaxmRNA表达的影响.方法:SW480细胞接种于6孔培养板中,24 h后分别以不同浓度(250、500、1000 μg/ml)的湿生扁蕾干预,继续培养24h后,提取每组总RNA,以GAPDH和β-actin基因为内参,采用RT-PCR法检测Bcl-2及Bax的mRNA表达水平.结果:Bcl-2 mRNA表达随湿生扁蕾浓度增加而减少,而Bax mRNA表达随湿生扁蕾浓度增加而增加,并呈现明显的剂量依赖关系.结论:藏药湿生扁蕾干预SW480细胞凋亡与其抑制Bcl-2的表达和促进Bax的表达有关.

Abstract: | Objective: Discuss the effect of Tibetan medicine gentianopsis paludosa for SW480 cell apoptosis regulatory factors Bcl -2mRNA and BaxmRNA. Methods: Inoculate SW480 cell in 6holes culture plate. Intervene in gentianopsis paludosa using different concentration (250, 500, 1000 μg/ml) after 24h. Continue to develop 24h, and extract each group total RNA. Use GAPDH and β - actin gene as internal reference. Then adopt RT - PCR assay to detect mRNA expression level of Bcl - 2 and Bax. Results : Bcl - 2mRNA expression are reducing with gentianopsis paludosa concentration increasing. While BaxmRNA expression is increasing with gentianopsis paludosa concentration increasing, and it presents the obvious dose dependency. Conclusion: Tibetan medicine gentianopsis paludosa control SW480 cells proliferation. It is connected with inhibition of Bcl -2 expression and promoting Bax expression. |
