| 摘 要： | 本文证明安定0.05mmol/L能显著抑制由氯化钙诱发的大鼠离体子宫体肌收缩。安定0.07mmol/L及硝苯啶0.028及0.33μmol/L使氯化钙的剂量反应曲线非平行性右移并降低其最大收缩反应,其PD_2′值分别为4.241及7.55、6.91。表明安定的钙拮抗作用弱于硝苯啶,安定对由高钾去极化所引起的大鼠离体子宫体收缩和依赖细胞内钙的收缩均有抑制作用,安定对子宫的抑制作用可能是通过对钙的拮抗作用所致。

| Abstract: | The present experiments demonstrated that diazepam (Diaz) 0.05 mmol/L significantly inhibited the contraction of rat isolated uterus induced by CaCl2. Diaz and nifedipine (Nif) shifted the dose-response curve of CaCl2 to the right non-paralleily and decreased the maximal response. The PD'2 value of Diaz and Nif was 4.24 and 7.55, 6.91 respectively, indicating that the calcium antagonistic effect of Diaz, was weaker than that of Nif. Diaz at the concentration of 0.05 mmol/L completely [inhibited the contraction of rat isolated uterus induced by the depolarization of high K+ (using Ca2+ 0.5 mmol/ L). Diaz and Nif also significantly inhibited intracellular Ca2+ dependent contraction. The inhibitory effects of Diaz on uterus were possibly mediated by calcium antagonistic effect. These effects offer an experimental basis for use of Diaz in obstetrical and gynecological clinical practice. |
