Determination of teicoplanin in serum by HPLC

Objective To establish an HPLC method for determination of teicoplanin in serum,providing the basis for monitoring clinical plasma concentrations and rational administration.Methods HPLC was performed on a C18 column（Wondasil,4.6 mm×250 mm,5 μm） with acetonitrile-0.05 mol/L monobasic potassium phosphate 28∶72 as mobile phase.The wavelength was 240 nm,and the flow rate was 1.0 mL/min,column temperature was 25 ℃.Results It showed good linearity in the concentration range of 4-48 μg/mL,and the limit of quantitation was 2 μg/mL,The average method recovery was 91.43% and the average extraction recovery was 88.40%.The inter-day RSD and intra-day RSD were both less than 6%.Conclusion The method was simple,quick and accurate,which could be used for the assay of teicoplanin in serum.
