肾炎防衰液对5/6肾切除大鼠肾组织FN表达的影响

目的探讨肾炎防衰液对5/6肾切除(5/6nephrectomize,5/6NT)大鼠肾组织纤维粘连蛋白(fibronectin,FN)表达的影响。方法选用SD雄性大鼠,建立5/6NT大鼠肾纤维化模型。随机分为8组:正常组、假手术组、模型组、苯那普利组、尿毒清组和肾炎防衰液低、中、高剂量组。采用免疫组织化学方法,以苯那普利、尿毒清为实验对照,检测肾炎防衰液对肾组织FN表达的影响。结果模型组肾组织FN强阳性表达,在系膜基质和肾小管基底膜出现大量棕黄色颗粒;苯那普利组、尿毒清组FN中等量表达,在系膜基质和肾小管基底膜出现中等量棕黄色颗粒;肾炎防衰液低、中、高剂量组肾组织FN轻度表达明显少于模型组,在系膜基质和肾小管基底膜出现少量棕黄色颗粒,并呈剂量依赖关系。结论肾炎防衰液抑制5/6NT大鼠肾组织FN的表达,可能是该方减缓肾小球硬化和肾纤维化发展的作用机制之一。

Influence of Shenyanfangshuai Solution on kidney FN expression in rats with 5/6 of nephrectomy

Objective To investigate the influence of Shenyanfangshuai Solution on the expression of kidney fibronectin(FN)in rats with 5/6 of nephrectomy.Method The model of renal fibrosis was established in SD male rats with 5/6 of nephrectomy,and then the rats were randomly divided into 8 groups:the normal group,sham-operation group,model group,benazepril group,Niaoduqing group and three Shenyanfangshuai Solution groups(in different doses).The influence of Shenyanfangshuai Solution on kidney FN expression was detected by immunohistochemistry method taking benazepril and Niaoduqing as experimental control.Result In the model group the kidney FN expression showed intense positive with a lot of brown depositions in the mesangial matrix and renal tubular basement membrane.In the benazepril group and Niaoduqing group the FN expression showed moderate positive with some brown depositions.In three Shenyanfangshuai Solution groups the FN expressions were much lower than those in the model group with a few brown depositions in a dose-reliable manner.Conclusion Shenyanfangshuai Solution inhibits the expression of kidney FN in rats with 5/6 of nephrectomy,which may be one of the mechanisms of Shenyanfangshuai Solution retarding the development of glomerulosclerosis and renal fibrosis.
