硫酸锌预处理对大鼠心脏缺血/再灌注损伤心肌超微结构的影响

摘    要:目的观察硫酸锌预处理对成年大鼠缺血再灌注损伤心肌的超微结构影响。方法将40只雄性SD大鼠随机分成4组(n=10),即正常组(N组)、缺血/再灌注组(C组)、硫酸锌预处理组(Zn组)、缺血预处理组(IPC组)。建立SD大鼠缺血再灌注损伤模型,用透射电子显微镜观察心肌组织的超微结构,并进行线粒体评分。结果 Zn组、IPC组、N组超微结构损伤程度及线粒体评分低于C组(P〈0.05)。Zn组、IPC组和N组心肌超微结构损伤及线粒体评分差异无统计学意义(P=1.00)。结论缺血/再灌注可对心肌细胞超微结构造成严重损伤,硫酸锌预处理和缺血预处理均可减轻缺血再灌注所致的心肌细胞超微结构损伤,由此可推测硫酸锌预处理可以产生心肌保护作用。

Abstract:Objective To observe the effects of zinc sulfate pretreatment on the in vitro cardiac ultrastructure of ischemia / reperfusion injury in adult rats. Methods Adult healthy male Sprague- Dawley rat hearts were isolated and subject to ischemia / reperfusion injury. This study was randomly divided into four groups( n = 10 in each group) : the normal group( N group),ischemia group( C group),zinc sulfate pretreatment group( zinc group),ischemic preconditioning( IPC group). The ultrastructure and mitochondria score of myocardial tissue in each group were observed by transmission electron microscope. Results The ultrastructure damage and mitochondria score of myocardial cells were attenuated in IPC and zinc groups compared with those in C group( P〈0. 05). No significant difference of the ultrastructure damage and mitochondria score of myocardial cells between IPC group and zinc group was shown( P = 1. 00). Conclusion Ischemia / reperfusion could lead to serious damage to ultrasture of myocardial cells and zine sulfate and ischemic preconditioning have similar protective effects on myocardium from ischemia / reperfusion injury in isolated rat hearts.
