双氢青蒿素对人肺腺癌细胞株A549抑制作用的实验研究

摘    要:目的观察双氢青蒿素对人肺腺癌细胞株A549的作用。方法采用四唑氮蓝(MTT)法测定不同浓度和时间双氢青蒿素对A549细胞的生长抑制作用;应用流式细胞术检测双氢青蒿素对A549细胞增殖和细胞周期的影响;应用透射电镜分析法检测双氢青蒿素对A549细胞凋亡的影响。结果双氢青蒿素对A549细胞有明显的体外增殖抑制作用,72 h的IC50值为580 nmol/L,且有明显的时间和剂量依赖关系;双氢青蒿素作用后G0/G1期细胞数增加;双氢青蒿素作用A549细胞后可以出现凋亡的形态学改变。结论双氢青蒿素对人肺腺癌A549细胞有生长抑制作用,其机制可能与诱导A549细胞周期阻滞和凋亡有关。

Abstract:Objective To observe the effect of dihydroartemisinin on the proliferation of human lung adenocarcinoma cell line A549 in vitro.Methods Cell proliferation in different concentration and time was determined by MTT(Methy thiazolyl tetrazolium) assay;The fluorescence flow cytometry was applied to detect the changes of cell proliferation and alteration of cell cycle phase;The apoptosis induction effects of A549 cells was studied by electron microscope observation.Results Dihydroartemisinin inhibited the proliferation of human lung adenocarcinoma cell line A549 cells in a time dependent and concentration-depended manner,after 72h of treatment,the IC50 value was 580 nmol/L;FCM assay indicated that cells in G0/G1 were increased after the dihydroartemisinin treatment;Electron microscope indicated that apoptotic change was observed in A549 cells after treatment with dihydroartemisinin.Conclusion Dihydroartemisinin has marked anticancer activity on human lung adenocarcinoma cell line A549 in vitro.The inhibition in vitro is related to arrest of cell cycle phase and apoptosis.
