| 摘 要： | 采用聚合酶链反应及基因工程技术，克隆卡介苗（ＢＣＧ）的主要分泌抗原Ａｇ８５Ｂ基因的５’端第９４～２１１的核苷酸的信号肽序列和构建重组人ＩＬ－２穿梭表达质粒，并用酶联免疫吸附法（ＥＬＩＳＡ）鉴定外源基因在ＢＣＧ中的表达。结果表明，构建的ＢＣＧ重组体能表达和分泌人ＩＬ－２。此将试用于临床治疗膀胱癌病人。

| Abstract: | Polymerase chain reaction and genetic engineering technique were used to clone the signal peptide sequence (position:94 to 211) of antigen 85-B in Mycobacterium bovis BCG and construct the recombinant E.coli-mycobacteria shuttle plasmoid of human interleukin-2(IL-2).Expression of the foreign gene in BCG was detected by enzyme linked immunosorbent assay. The results suggest that the constructed recombinant BCG(rBCG) strain produce and secrete human IL-2 and it will be used for the treatment of superficial bladder cancer. |
