Effect of pseudo-allergic drug on memory after osplenectomy in rats

Objective To observe the effect of pseudo-allergic drug on memory after osplenctomy in rats.Methods Healthy male sprague dawley(SD)rats,a total of 48,aged 22 months and weighing(350±50)g,were purchased from the Henan Experimental Animal Centre,and were randomized into 4 groups:anesthesia without surgery group(group A),anesthesia and surgery group(group B),anesthesia and allergy group(group C),anesthesia and surgery with allergy group(group D).The rats in each group were numbered and given Morris Water Maze Training,and the training targets were as follows:6 rats with an escape latency of about 20 s and rats in A,B,C and D groups were trained in escape latency.When the training latency was about 20 seconds,six rats were assigned to A1,B1,C1,D1 groups,the remaining 6 rats in each group continued to train until the escape latency was about 10 seconds and were grouped as A2,B2,C2 and D2.After general anesthesia with Fentanyl(20μg/kg)+droperidol(500μg/kg)in each group,C-C motif chemokine 11(CCL11),histamine and complement complement 5B-9 complex containing s protein(SC5b-9)were tested for the first blood collection(submandibular vein blood collection).The operation model(splenectomy)and(or)allergy model[injection of 5%cremophor polyoxyethylene castor oil(CrEL)via the rat tail vein]were established according to the requirements of experimental grouping.At last,CCL11,histamine and complement activated fragment SC5b-9 were detected by the second submandibular venous blood sampling in anesthetized rats,the level of expression was detected by enzyme linked immunosorbent assay(ELISA),and water maze test was carried out on the 1st,3rd and 5th day after intervention to observe the change of escape latency.Results There was no significant difference between the four groups in subsection training level of escape latency(F=0.040,P>0.05;F=0.397,P>0.05).There was significant difference in escape latency between the control group A1 and group B1(22.8±3.1 vs.18.1±2.9,20.4±2.7 vs.16.7±2.1,16.7±2.8 vs.13.2±2.2)at 1st,3rd and 5th day after intervention(t=2.712,P<0.05,t=2.649,P<0.05,t=2.407,P<0.05).After intervention,there was significant difference in the escape latency between group A1 and group C1 at 1st,3rd and 5th day(24.7±5.1 vs.18.1±2.9,23.9±3.5 vs.16.7±2.1,17.7±3.9 vs.13.2±2.2)(t=2.755,P<0.05,t=4.321,P<0.01,t=2.462,P<0.05),and between group A1 and group D1(26.4±3.4 vs.18.1±2.9,25.8±3.0 vs.16.7±2.1,24.1±3.8 vs.13.2±2.2)(t=4.549,P<0.01;t=5.239,P<0.01;t=6.081,P<0.01).There was significant difference between control group A2 and group B2 in escape latency(13.4±2.5 vs.9.5±3.2,12.2±2.1 vs.9.4±2.0)at 1st and 3rd days after intervention(t=2.353,P<0.05,t=2.365,P<0.05),with no significant difference in escape latency(9.8±1.7 vs.9.2±1.4)at 5th day(t=0.667,P>0.05),while there was significant difference between control group A2 and group C2 in escape latency(16.9±3.1 vs.9.5±3.2,15.2±3.7 vs.9.4±2.0)at 1st and 3rd days after intervention(t=4.068,P<0.01,t=3.378,P<0.01),with no significant difference in escape latency(11.4±2.1 vs.9.2±1.4)in the group C2 at 5th day(t=2.135,P>0.05),between group D2 and group A2(18.9±3.3 vs.9.5±3.2,18.2±3.0 vs.9.4±2.0,15.1±2.9 vs.9.2±1.4)at 1st,3rd and 5th day(t=5.009,P<0.01;t=5.978,P<0.01,t=4.488,P<0.01).CCL11,histamine and complement SC5b-9 in the first blood collection of the four groups had no significant difference(F=0.031,P>0.05;F=1.087,P>0.05;F=0.826,P>0.05).At the second submandibular venous blood sampling,there was no significant difference between group B and group A[(135.87±19.74)vs.(125.76±19.18)ng/L;(1.90±0.23)vs.(1.88±0.17)mg/L;(30.07±4.25)vs.(28.55±3.74)μg/L,t=1.272,P>0.05;t=0.242,P>0.05;t=0.930,P>0.05];there was significant difference between group C and group A[(328.77±42.95)vs.(125.76±19.18)ng/L;(2.53±0.29)vs.(1.88±0.17)mg/L;(39.88±5.64)vs.(28.55±3.74)μg/L](t=14.951,P<0.01;t=6.595,P<0.01;t=5.799,P<0.01),and between group D and group A[(471.49±44.58)vs.(125.76±19.18)ng/L;(3.38±0.34)vs.(1.88±0.17)mg/L;(48.12±5.13)vs.(28.55±3.74)μg/L](t=24.678,P<0.01;t=13.670,P<0.01;t=10.678,P<0.01);CThere was significant difference between group C and group D(t=7.987,P<0.01;t=6.589,P<0.01;t=3.733,P<0.01).Conclusion The occurrence of pseudo-allergic drug during operation has great influence on the short-term memory and stable memory of rats,which may be related to CCL11.
