宜昌地区人群TLR4受体Asp299Gly、Thr399Ile位点多态性研究

目的研究TLR4受体Asp299Gly、Thr399Ile位点多态性在宜昌地区健康汉族人群中的分布。方法采用聚合酶链反应-限制性片段长度多态性分析技术(PCR—RFLP)法对156例宜昌地区无血缘关系的汉族健康人,进行TLR4受体Asp299Gly、Thr399Ile两位点的基因型检测。结果宜昌地区无血缘关系的汉族健康人TLR4受体基因Asp299Gly位点男女Asp／Asp、Asp／Gly和Gly／Gly的基因型频率分别为0．818,0．148,0．034和0．824,0．147, 0．029:Thr399Ile位点男女Thr／Thr、Thr／Ile和Ile／Ile的基因型频率分别为0．977,0．023,0．000和0．941,0．044, 0．015。与英国、法国等国人群相比,该位点多态性不存在显著性差异。TLR4受体Asp299Gly、Thr399Ile两位点的基因型男女之间也无显著性差异(X2=0．028,P=0．986)(X2=1．894,P=0．388)。结论宜昌地区健康汉族人群存在TLR4受体Asp299Gly、Thr399Ile两位点的基因多态性,并在不同种族和性别间的分布不存在显著性差异。

The Asp299Gly and Thr399Ile Polymorphisms in the Toll- like Receptor 4 of Han Nationality in Yichang

Objective This study was to research some polymorphisms in the Toll -like receptor 4 (TLR4) of Han nationality in Yichang. Methods DNA were obtained from a group of healthy subjects. The subjects were genotyped for the Asp299Gly and Thr399lle Polymorphisms in the Toll - like Receptor 4 by Restriction fragment length polymorphism polymerase chain reaction (PCR -RFLP). Results The genotype frequencies were 0. 818 for Asp/Asp,0.148 for Asp/Gly and 0.034 for Gly/Gly in male; the genotype frequencies were 0. 824 for Asp/Asp,0. 147 for Asp/Gly and 0.029 for Gly/Gly in female of Han nationality in Yichang. The genotype frequencies were 0.977 for Thr/Thr,0.023 for Thr/lle and 0.000 for lie/lie of healthy subjects in male; the genotype frequencies were 0. 941 for Thr/Thr , 0. 044 for Thr/lle and 0. 015 for He/lie of healthy subjects in female of Han nationality in Yichang. No significant difference in the distribution of Asp299Gly and Thr399lle polymorphism was found in different sex and races. Conclusion These data suggested that some polymorphisms in the Toll -like receptor4 (TLR4) of Han nationality in Yichang. However, no significant difference in the distribution of Asp299Gly and thr399lle polymorphism was found in different sex and races.
