槲皮素及阿司匹林对血小板与中性粒细胞相互作用的影响

摘    要:采用血小板聚集及血小板与中性粒细胞形成玫瑰花结试验探讨槲皮素及阿司匹林对血小板与中性粒细胞相互作用的影响. 结果表明,家兔中性粒细胞可抑制ADP诱导的血小板聚集,槲皮素(0.3-100 μmol·L-1)及阿司匹林(0.4-3.3 mmol·L-1)可增加中性粒细胞对血小板的抑制作用. 当中性粒细胞被乙酸肉豆寇佛波醇(PMA 100 μg·L-1)激活时,此抑制作用消失. 槲皮素 (3 μmol·L-1)可对抗PMA激活中性粒细胞. 即使无中性粒细胞存在,阿司匹林亦可抑制血小板的聚集,但单独槲皮素却不抑制血小板聚集. 槲皮素(3-300 μmol·L-1)及阿司匹林(0.4-3.3 mmol·L-1)可浓度相关性地抑制凝血酶激活的血小板与中性粒细胞的粘附.

Abstract:Platelet aggregometry and platelet/neutrophil rosetting assay were used to investigate neutrophils mediated anti-platelets aggregation and platelet/neutrophil adhesion, as well as the effect of quercetin and aspirin on platelet-neutrophil interaction. Unactivated rabbit neutrophils inhibited ADP- induced platelet activation; both aspirin (0.4-3.3 mmol·L-1) and quercetin (0.3-100 μmol·L-1) increased this inhibitory effect of neutrophils. When the neutrophils were stimulated by phorbol myristate acetate (PMA, 100 μg·L-1), the inhibitory effect was disappeared. Quercetin (3 μmol·L-1) protected neutrophils from activation by PMA. Aspirin also inhibited platelet activation even in the absence of neutrophils but quercetin did not. Both quercetin (3-300 μmol·L-1) and aspirin (0.4-3.3 mmol·L-1) caused a concentration- dependent inhibition of thrombin activated platelets binding to neutrophils.
