MMP-9，TIMP-1蛋白在肺癌患者血清中表达水平及与近期化疗疗效的关系

目的：本研究通过检测MMP-9，TIMP-1在肺癌患者血清中的表达，研究二者与临床病理特征之间的关系，并探讨二者在肺癌近期化疗疗效评估中的价值。方法3组研究对象为肺癌组70例，支气管、肺良性病变组31例，健康对照组18例。其中37例肺癌患者行全身化疗，2个化疗周期后，根据影像学评估疗效。应用酶联免疫吸附法（ ELISA）检测血清含量。结果肺癌组血清MMP-9，TIMP-1水平明显高于健康对照组及良性病变组，差异有显著性（P＜0．05）。化疗前血清MMP-9，TIMP-1水平与年龄、性别、病理类型、分化程度无相关性，但与肿瘤大小、纵膈淋巴结转移及远处转移有相关性（P＜0．05）。肺癌组患者在化疗前血清中MMP-9与TIMP-1水平正相关，Spearman’s相关系数为0．877。肺癌组化疗有效者MMP-9水平降低，差异有统计学意义，TIMP-1水平虽有下降，但无统计学意义。病情稳定及进展者水平无明显变化。结论 MMP-9，TIMP-1与肺癌的浸润、转移密切相关，检测血清MMP-9，TIMP-1水平有助于指导临床肺癌诊断及评价疗效。

The Serum Expression of MMP-9 and TIMP-1 Protein in Lung Cancer Patients and Their Relationship with the Short-term Chemotherapeutic Efficacy

Objective To explore the relationship between MMP-9 and TIMP-1 serum expressions and clinical pathological fea-tures of lung cancer patients ,to discuss the value of application for the changes of their serum expression in evaluation of recent chemotherapy response.Methods The concentrations of serum MMP-9 and TIMP-1 were measured by ELISA respectively in 70 patients with lung cancer and 31 patients with benign lung diseases as well as 18 healthy persons served as control .There were 37 patients in lung cancer group treated by chemotherapy and the chemotherapy effect was evaluated after 2 cycles.Meanwhile,to detect the serum level of MMP-9 and TIMP-1.Re-sults The level of expression of MMP-9 and TIMP-1 in serum of lung cancer was significantly higher than that of healthy and benign respira -tory disease.There were no significant differences among the MMP-9,TIMP-1 expression that before the treatment and age ,sex,histological type and differentiation.There was statistical relationship between expression of MMP-9,TIMP-1 and tumor size,lymph node metastasis and distant metastasis.There was a positive correlation（r=0.877） between MMP-9 and TIMP-1 in the serum of lung cancer patients before the treatment .After chemotherapy ,the serum levels of MMP-9 were decreased significantly in the CR and PR patients ,at the same time ,although the expression of TIMP-1 was decreased ,there was no significant difference .In non-responders before and after chemotherapy there was no significant difference in serum MMP-9 and TIMP-1 levels.Conclusion This study has confirmed that MMP-9,TIMP-1 expressions are relat-ed to the infiltration and metastasis of lung cancer .And the variation of serum MMP-9 and TIMP-1 levels has clinical significance in diagno-sing the lung cancer and estimating the chemotherapeutic efficacy .
