白藜芦醇苷对脑缺血再灌注模型大鼠的保护作用

摘    要:目的:研究白藜芦醇苷对脑缺血再灌注模型大鼠的保护作用。方法:采用栓线法制备大鼠急性局部脑缺血再灌注损伤模型。140只SD大鼠随机分为假手术(等容生理盐水)组、模型(等容生理盐水)组、依达拉奉(6mg/kg)组与白藜芦醇苷①、②、③、④(40、20、10、5mg/kg)组。再灌注1h后舌下静脉给药。给药24h后断头取脑行TTC染色,测定脑梗死体积百分比与脑组织含水量。180只sD大鼠随机分为假手术(等容生理盐水)组、模型(等容生理盐水)组、依达拉奉(6mg/kg)组与白藜芦醇苷高、中、低剂量(20、10、5mg/kg)组。再灌注3h后舌下静脉给药,每天1次,连续8d。在给药第2、4、6、8天时称定体质量并计算变化率,进行姿势反射、肢体不对称实验。结果:与假手术组比较,模型组大鼠脑梗死体积百分比与脑组织含水量显著增加(P〈0.01);与模型组比较,白藜芦醇苷①、②、③组大鼠脑梗死面积百分比与脑组织含水量显著减少(P〈0.01或P〈0.05)。与假手术组比较,模型组大鼠体质量变化率显著增加(P〈0.01),姿势反射、肢体不对称实验评分显著升高(P〈0.01);与模型组比较,白藜芦醇苷高剂量组大鼠在给药第2、6、8天时姿势反射实验评分显著降低(P〈0.01),白藜芦醇苷高、中、低剂量组大鼠在给药第4天时姿势反射实验评分显著降低(P〈0.01或P〈0.05),白藜芦醇苷高剂量组大鼠在给药第2、6天时肢体不对称实验评分显著降低(P〈0.01或P〈0.05),白藜芦醇苷高、中、低剂量组大鼠在给药第4天时肢体不对称实验评分显著降低(JP〈O.01或P〈0.05)。结论:白藜芦醇苷对脑缺血再灌注模型大鼠具有一定保护作用。

Protective Effects of Resveratrol on Cerebral Ischemia-reperfusion Model Rats

Abstract:OBJECTIVE: To study the effects of resveratrol in rats after focal cerebral ischemia-reperfusion injury. METHODS: Focal cerebral ischemia-reperfusion injury model was induced by thread occlusion method. 40 SD rats were randomly divided into sham operation group (constant volume of normal saline), model group (constant volume of normal saline), edaravone group (6 mg/kg) and first, second, third and forth resveratrol groups (40,20, 10,5 mg/kg). Those were given medicines via sublingual vein 1 h after reperfusion. Rats were decapitated and cerebral TTC staining were performed 24 h after medication, the volume of cere- bral infraction and water content were determined. 180 SD rats were randomly divided into sham operation group (constant volume of normal saline), model group (constant volume of normal saline), edaravone group (6 mg/kg) and resveratrol high-dose, medi- um-dose and low-dose groups (20, 10,5 mg/kg). They were given medicines via sublingual vein 3 h after perfusion, once a day, for consecutive 8 days. The body weight of rats was determined on 2nd, 4th, 6th and 8th day after medication and the change rate was also calculated. Postural reflex test and limbs asymmetry test both were performed. RESULTS: Compared with sham operation group, the percentage of cerebral infraction volume and water content of cerebral tissue were increased significantly in model group (P〈0.01). Compared with model group, the percentage of cerebral infraction volume and water content of cerebral tissue were de- creased significantly in first, second and third resveratrol groups(P〈0.01 or P〈0.05). Compared with sham operation group, the change rate of body weight was increased significantly in model group(P-〈0.01 ), and postural reflex test and limbs asymmetry test score were increased significantly(P〈0.01). Compared with model group, postural reflex test score of resveratrol high-dose group was decreased significantly on 2nd, 6th and 8th day of medication (P〈0.01) ; postural reflex test scores of resveratrol high-dose, medium-dose and low-dose groups were decreased significantly on 4th day of medication (P〈0.01 or P〈0.05). The limbs asymme- try test score of resveratrol high-dose group was decreased significantly on 2nd and 6th day of medication(P〈0.01 or P〈0.05) ; limbs asymmetry test score of resveratrol high-dose group, medium-dose group and low-dose group was decreased significantly on4th day of medication (P〈0.01 or P〈0.05). CONCLU- SIONS: Resveratrol has protective effects against cerebral isch- emia-reperfusion injury.
