改良乌头汤减毒增效比较研究

摘    要:目的:研究不同配伍下乌头汤毒性生物碱成分的变化及其镇痛作用、急毒性。方法:采用高效液相色谱法测定不同配伍下乌头汤中苯甲酰乌头原碱、苯甲酰次乌头原碱、苯甲酰新乌头原碱的含量变化;采用醋酸致小鼠扭体模型,比较研究不同配伍下乌头汤的镇痛作用;研究不同配伍下乌头汤的急毒性。结果:与制川乌单煎比较,改良乌头汤(乌头原汤加防风、黑豆)中苯甲酰乌头原碱、苯甲酰次乌头原碱、苯甲酰新乌头原碱含量分别降低了73.09%、68.01%、74.53%;17.50、8.75、4.35g/kg改良乌头汤与12.50、6.25、3.13g/kg乌头原汤可明显减少醋酸致小鼠20min内扭体次数(P〈0.01或P〈0.05),17.50g/kg改良乌头汤与12.50、6.25g/kg乌头原汤可显著延长扭体潜伏期(P〈0.05);改良乌头汤无急毒性反应。结论:乌头原汤配伍防风、黑豆能减少乌头原汤毒性生物碱成分的含量,鸟头原汤与改良乌头汤在镇痛药效方面无明显差异。

Abstract:OBJECTIVE: To study the change of toxic alkaloid in different compatibility of Wutou decoction and its pharmaco- dynamics and acute toxicity. METHODS: HPLC was applied to determine the contents of benzoylmesaconine, benzoylaconine, ben- zoylhypacoitine of Wutou decoction. Analgesic actions of different compatibility of Wutou decoction were compared by the methods of glacial acetic acid induced writhing test; the acute toxicity of it was studied. RESULTS: Compared with Processed Radix Aconiti Praeparat extract, the contents of benzoylaconine, benzoylhypacoitine, benzoylmesaconine decreased by 73.91% , 68.01, 7.53 in modified Wutou decoction(Wutou decoction combined with Saposhnikovia. divaricata and Glycine max)respectively; modified Wut- ou decoction (17.50,8.75,4.35 g/kg)and Wutou decoction (12.50,6.25,3.13 g/kg)could reduce writhing response within 20 min sig- nificantly(P〈0.01 or P〈0.05). modified Wutou decoction (17.50 g/kg)and Wutou decoction (12.50,6.25 g/kg )could extend the writhing latency of mice, but there was no significant difference in analgesic action. No acute toxic reaction was found.CONCLU- SIONS: Wutou decoction combined with S. divaricata and G. max could reduce the content of toxic alkaloid, but the analgesic ef- fect has no changes.
