急性胰腺炎时胰腺细胞亚细胞器的功能变化及栀子的影响

取SD大鼠18只(200～250g),随机均分三组:正常、胰炎+盐水及胰炎+栀子。用1.5％DCA-Na诱发急性胰腺炎实验模型,术前1小时,术后4小时及细胞学实验前1小时分别给栀子煎剂或生理盐水,正常动物不给处理。术后24小时用差速离心制备溶酶体及线粒体,以酸性磷酸酶和琥珀酸脱氢酶分别作为标志酶检测它们的功能。实验揭示盐水组的酸性磷酸酶显著高于正常,琥珀酸脱氨酶则显著降低;而栀子组则与正常近似。结果提示胰炎时胰腺细胞的氧化代谢和溶酶体功能发生异常,而用栀子治疗可减轻之。

EFFECT OF GARDENIA JASMINOIDES ELLIS ON THE FUNCTIONAL CHANGE OF PANCREATIC SUBCELLULAR ORGANS (ABSTRACT)

18 SD rats weight 200-250g were divided randomly into 3 groups: normal,pancreatitis + saline and pan creatitis + GJE. The experimental acutepancreatitis model was induced with 1.5% DCA-Na injection, andtreated with GJE decoction or saline 1h pre-and 4h post-operationand also 1h before cellular examination. The normal animals wereuntreated. 24h after operation. The lysosome and metochondria wereprepared with density gradient centrifugation and acid phosphatase(CAP) and succinic dehydrogenase (SDH) were measured as markedenzyme respectively to evaluate their function. It was demonstratedthat the CAP raised and SDH reduced significantly than the normalin the saline group, but nearly as same as normal in GJE group. Itwas suggested that the oxidative metabolism and lysosomal function ofpancreatic cell were disorder in pancreatitis, and which would bereduced by GJE treated.
