摘    要:目的:建立可用于药物筛选的体外蛋白质非酶糖基化模型。方法:37℃孵育1周后用流动注射法于波长(λ:Ex/Em):370/440nm处测定糖基化终产物(AGEs)。结果:反应1周后,反应体系内3-脱氧葡萄糖醛酮(3.DG)浓度与AGEs的吸收度值呈线性关系,y=22.946x+22.846;r^2=0.9985。结论:随着各反应体系中底物3-DG浓度的增加及反应时间的延长,蛋白质非酶糖基化反应增强3-DG为底物与牛血清白蛋白组成反应系统,能较可靠地反映AGEs的生成变化,有望成为体外蛋白质非酶糖基化的1种新模型。

Establishment of the Reaction System for Advanced Glycation End Products Induced by 3-deoxyglucosone in vitro

Abstract:Objective:To establish the reaction of nonenzymatic glycation of protein.Methods:BSA(bovine serum albumin)was incubated with 3-DG(3-deoxyglucosone)for one week at 37℃in vitro.AGEs(advanced glycation end products)was estimated by FIA atλ370/440 nm(excitation/emission).Results:AGEs were increased accompanying with the increasing of 3-DG,showing positive linear relations(v=22.946x+22.846;r~2=0.998 5).Conclusions:The present study showed that the reaction of nonenzymatic glycation of protein was reinforced with the concentration increasing and time lengthening of 3-deoxyglucosone.Furthermore,our results supported the notion that the reacting systems,composing 3-deoxyglucosone incubatirig with bovine serum albumin,might become a novel model for researching AGEs formed in vitro due to their characters of monitoring the variances of AGEs sensitively.
