粒径和MePEG相对分子质量对隐形纳米粒固着水层厚度和表面MePEG链密度的影响

摘    要: 目的考察粒径和单甲氧基聚乙二醇(methoxypolyethyleneglycol,MePEG)相对分子质量对重组人肿瘤坏死因子(recom-binant human tumor necrosis factor-α,rHuTNF-α)隐形纳米粒固着水层厚度(fixed aqueous layer thickness,FALT)和表面MePEG链密度的影响。方法复乳法制备3种不同粒径(80,170和240 nm)和表面3种不同相对分子质量MePEG(Mr=2 000,5 000,10 000)修饰的重组人肿瘤坏死因子聚乙二醇化聚十六烷基氰基丙烯酸酯(poly,methoxypolyethyleneglycol cyanoacrylate-co-n-hexadecyl cyanoacrylate,PEG-PHDCA)隐形纳米粒。测定不同纳米粒的FALT和表面MePEG链密度,并加以比较。结果粒径相同时,随着MePEG相对分子质量的增加,FALT增大,表面MePEG链密度减小;MePEG相同时(Mr=5 000),随着粒径的减小,FALT增加,表面MePEG链密度增大。结论实验范围内,粒径和MePEG相对分子质量对重组人肿瘤坏死因子隐形纳米粒的FALT和表面MePEG链密度有显著影响。

Abstract:OBJECTIVE To investigate the influence of particle size and methoxypolyethyleneglycol(MePEG) molecular weight on fixed aqueous layer thickness(FALT) and surface MePEG chain density of stealth nanoparticles.METHODS Three sizes(80,170 and 240 nm) of poly(methoxypolyethyleneglycol cyanoacrylate-co-n-hexadecyl cyanoacrylate)(PEG-PHDCA) nanoparticles loading rHuTNF-α were prepared at different MePEG molecular weights(Mr=2 000,5 000,10 000) using the double emulsion method.The physicochemical properties including FALT and surface MePEG chain density were examined and compared.RESULTS With the same size,FALT was increased but surface MePEG chain density were decreased as MePEG molecular weight were increased.With the same MePEG(Mr=5 000) coating,both FALT and surface MePEG chain density were increased as particle size was decreased.CONCLUSION Within the experimental limits,particle size and MePEG molecular weight had dramatic influence on FALT and surface MePEG chain density of rHuTNF-α-loaded stealth nanoparticles.
