化浊通脉散对颈动脉粥样硬化家兔凝血机制变化和eNOS表达的影响

摘    要:目的研究化浊通脉散对颈动脉粥样硬化家兔凝血机制变化及动脉血管内皮的影响,并对其发生机制进行初步探讨。方法 40只新西兰大耳白兔采用高脂饮食的实验方法建立动脉粥样硬化模型,随机分为4组:化浊通脉散组、阿司匹林组、高脂饮食组、空白对照组。各组分别测定药物干预前后血浆凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、血浆纤维蛋白原(Fg)、血浆凝血酶时间(TT)水平。实验结束后应用免疫组织化学方法观察内皮型一氧化氮合成酶(eNOS)的表达。结果化浊通脉散有延长PT、APTT、TT时间及降低Fg含量的作用(P0.01),并可提高颈动脉粥样硬化血管内膜eNOS的表达。结论化浊通脉散具有抗凝及降纤作用,并可以使血管内膜eNOS活性增强,因此可能具有保护动脉血管内膜,延缓动脉粥样硬化形成的作用。

Abstract:Objective To study the impacts of huazhuotongmai powder on coagulation changes and artery endothelium in rabbits with carotid atherosclerosis and explore primarily its mechanism.Methods Atherosclerosis models were set up in 40 New Zealand white rabbits by high-fat diet and were randomly divided into huazhuotongmai powder group,aspiring group,high-fat diet group and control group.Before and after drug intervention,plasma coagulation ezymogen time(PT),activated partial thromboplastin time(APTT),fibrinogen(Fg),thrombin time(TT)levels were determined separately in each group.At the end of experiment,immunohistochemistry assay was adopted to observe the expression of endothelial nitric oxide synthase(eNOS).Results Huazhuotongmai powders could prolong the time of PT,APTT and TT,reduce Fg content(P0.01)and increase eNOS expression in carotid atherosclerotic vascular endothelium.Conclusion Huazhuotongmai powder acts on anticoagulation,reduces fibrosis and increases eNOS activity of arterial endothelium.Hence,the drug can protect arterial endothelium and delay the formation of atherosclerosis.
