缺氧及氧化损伤后GEC粘附分子表达与粘附性改变

摘    要:目的:探讨缺氧、氧化损伤对培养人肾小球内皮细胞(GEC)粘附分子表达及GEC与PMN粘附力的影响。方法:采用化学缺氧、氧化损伤模型,免疫荧光观察GEC粘附分子P、E选择素和ICAM-1的表达,微管吸吮直接测定单个GEC、PMN细胞间粘附力的改变。结果:缺氧、氧化损伤均可导致GEC细胞、P、E选择素和ICAM-1的表达增强,以ICAM-1最明显,同时GEC、PMN间粘附力增强,加用ICAM-1抗体可显著抑制这一粘附力的增加。结论:缺氧、氧化损伤后GEC粘附分子表达增强,GEC、PMN细胞间粘附力的改变与GEC粘附分子变化密切相关。

Abstract:Objective To explore the effects of anoxia/oxidative stress on the expressions of adhesion molecules on glomerular endothelial cells (GECs) and its adhesion properties. Methods Chemical anoxia/oxidative stress models of GECs were established, and the expressions of P selectin, E selectin and ICAM 1 in them were studied with immunofluorescence. Micropipette aspirate techniques were used to measure the adhesive forces between GECs and PMNs. Results Anoxia/oxidative stress increased the expression of P selectin, E selectin and ICAM 1, with the last one most obvious. Adhesive forces between GECs and PMNs increased simultaneously. Antibody to ICAM 1 inhibited the adhesive forces between GECs and PMNs markedly. Conclusion The expressions of adhesion molecules on GECs increased after anoxia/oxidative stress, and these changes are related to the changes of adhesive forces between GECs and PMNs.
