环维黄杨星D对去甲肾上腺素诱导大鼠心肌细胞凋亡影响

摘    要:目的研究环维黄杨星D(CVB-D)对去甲肾上腺素(NE)诱导乳鼠心肌细胞损伤凋亡的影响。方法取SD大鼠乳鼠心肌细胞做原代培养,20μmol.L-1NE诱导心肌细胞凋亡模型,Giemsa染色TUNEL、DNA-Ladder凝胶电泳检测心肌细胞凋亡,免疫组化法检测caspase-3蛋白的表达,检测细胞LDH的外漏率,线粒体琥珀酸脱氢酶(MSDH)的活力,观察CVB-D的保护作用。结果模型组Giemsa染色形态学上出现典型的凋亡特征;DNA Ladder凝胶电泳出现典型的梯状条带;TUNEL染色法测定的凋亡率增高;Caspase-3的阳性表达率增高,细胞LDH的外漏率增加,MSDH活力降低。CVB-D明显的降低了细胞LDH的外漏率,提高了MSDH的活力,对心肌细胞的凋亡具有明显的保护作用。结论 CVB-D对心肌细胞凋亡和细胞的能量代谢障碍具有显著的保护作用。

Abstract:Objective To investigate the cardio-protective effect of cyclovirobuxinum D(CVB-D) on primary culture of cardiac myocyte treated with noradrenaline.Methods After adding CVB-D,the primary culture of cardiac myocyte was injured by 20μmol·L-1 NE.The activity of lactic dehydrogenase(LDH) and mitochondrion succinic dehydrogenase(MSDH) were assayed.The percentage of cardiac myocyte apoptosis was observed by Giemsa staining、 TUNEL、DNA-Ladder and immunohistochemistry was used for detection of Caspase-3 protein.Results CVB-D significantly decreased the activity of LDH in culture supernatant,increased the activity of MSDH.Conclusion CVB-D can alleviate the disturbance of energy metabolism and decrease the percentage of apoptosis of cardiac myocyte treated with noradrenaline.It has a significant protective effect on myocardial apoptosis.
