赤雹根总皂苷镇痛作用机制研究

摘    要:目的探讨赤雹根总皂苷(TSTD)镇痛作用与阿片受体、NO及PGE2的关系,为其临床应用提供实验依据。方法采用小鼠热板法实验,观察纳洛酮、L-精氨酸(L-Arg)和左旋硝基精氨酸甲酯(L-NAME)对赤雹根总皂苷镇痛作用的影响,探讨其镇痛作用与阿片受体及NO含量的关系;通过观察TSTD对疼痛模型小鼠组织中PGE2含量的影响,探索其镇痛作用与PGE2的关系。结果 TSTD组痛阈显著高于模型组(P<0.01);TSTD组与纳洛酮+TSTD组痛阈差异无显著统计学意义(P>0.05);L-Arg+TSTD组痛阈显著低于TSTD组(P<0.01)。TSTD+L-NAME组痛阈高于TSTD组。TSTD组小鼠血清和脑组织中NO的含量显著低于NS对照组(P<0.05),TSTD+L-Arg组NO含量高于TSTD组(P<0.05),TSTD+L-NAME组NO含量低于TSTD组(P<0.05)。TSTD组动物腹腔液和血清中PGE2的含量低于NS对照组(P<0.01)。结论 TSTD镇痛作用与阿片受体无关,与抑制NO和PGE2的合成或释放有关。

Abstract:Objective To study the mechanism of analgesic effect of total saponin from the root of Thladiantha Dubia Bunge(TSRT).Methods To investigate the relationship between analgesic effect and opium receptor,levels of No and PGE2.The effects of Naloxone,L-Arg and L-NAME on the analgesic effect of TSRT were evaluated by tail-immersion test(50℃)in mice.The contents of NO and PGE2 in serum and brain tissue of animals were measured.Results The TSRT markedly heightened pain threshold in mice(P<0.01 vs NS group) and its analgesic effect was not antagonized by Naloxone.Analgesic effect of TSTR was partially antagonized by ip L-Arg.The pain threshold in TSTD+L-Arg group was less than that of TSTE group(P<0.05),and which could be markedly enhanced by ip L-NAME-TSTD significantly reduced the contents of NO and PGE2 in serum or brain tissue of model animals(P<0.01 vs NS group).The content of NO was much higher in TSTD + L-Arg group than that in TSTD group(P<0.01),but it was much lower in TSTE + L-NAME group than that in TSTD group.Conclusion The analgesic of TSTD can not be mediated by opium receptor,but it can suppress the synthesis or releases of excessive NO and PGE2.
