果糖二磷酸钠对2型糖尿病大鼠周围神经传导功能的影响

摘    要:目的:研究果糖二磷酸钠(FDP)对2型糖尿病(T2DM)大鼠周围神经病变的影响。方法:首先建立T2DM大鼠模型,将实验动物分成4组,正常对照组、模型对照组、FDP高剂量治疗组(5mL.kg-1.d-1,i.p.)和FDP低剂量治疗组(2mL.kg-1.d-1,i.p.),每组8只。8周末,测定大鼠尾运动神经传导速度(MCV)、感觉神经传导速度(SCV),H反射中H与M波的潜伏期,血清血糖(FBG)与超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)的变化。结果:①与正常组比较,FDP干预前,T2DM大鼠FBG、血清胰岛素(FINS)水平显著升高,胰岛素敏感指数(ISI)显著降低(P〈0.01);②FDP干预后,与正常组比较,模型对照组FBG、FINS水平显著升高,ISI、SOD、NOS和MCV、SCV显著下降,H与M波潜伏期显著延长(P〈0.01);③与模型对照组比较,FDP干预后,高剂量组大鼠FBG、FINS水平显著降低,ISI、SOD、NOS、MCV和SCV显著升高,H与M波潜伏期显著缩短(P〈0.01);低剂量组大鼠FBG显著降低,SOD、NOS、MCV和SCV显著升高,M波潜伏期显著缩短(P〈0.01或P〈0.05);④与低剂量治疗组比较,高剂量组大鼠FBG、FINS水平显著降低,ISI、SOD、NOS、MCV和SCV显著升高,H与M波潜伏期显著缩短,(P〈0.01或P〈0.05)。结论:FDP对糖尿病大鼠周围神经病变具有一定的治疗作用,其机制可能与FDP降低糖尿病大鼠FBG和提高机体抗氧化能力有关。

Abstract:Objective:To study the effects of fructose sodium diphosphate(FDP) on peripheral nerve conduction function with type 2 diabetes mellitus(T2DM) rats.Methods:Male Wistar rats were developed the model of T2DM,these rats were divided into four groups(n=8):normal control group,model control group,high dose group(5mL·kg-1·d-1,i.p.),low dose group(2mL·kg-1·d-1,i.p.).The treatment group rats were received FDP for 8 weeks,the following parameters were measured on:the fasting blood sugar(FBG),SOD,NOS,insulin and the sensitivity index(FINS,ISI),movement nerve conduction velocity(MCV),sensory nerve conduction velocity(SCV),incubation period of H wave and M wave in the H-reflex.Results:①Compared with normal control group,FBG,FINS and the incubation period of H wave and M wave in the model control group were significantly increased;ISI,SOD,NOS,MCV and SCV were significantly decreased(P〈0.01);②Compared with model control group,in the high-dosage groups,the FBG,FINS and incubation period of H wave and M wave were significantly decreased;ISI,SOD,NOS,MCV and SCV were significantly increased(P〈0.01);in low doses groups,the FBG,SOD,NOS,and incubation period of M wave were significantly decreased;the MCV and SCV were significantly increased(P〈0.05).③Compared with low doses groups,in the high-dosage groups,the FBG,FINS and incubation period of H wave and M wave were significantly decreased;ISI,SOD,NOS,MCV and SCV were significantly increased(P〈0.01 or P〈0.05).Conclusion:FDP may prevent and treat the diabetic peripheral neuropathy in T2DM rats,the FBG decrease and the antioxidation enhancement may play an important role.
