甘露醇与尼莫地平联合应用对大鼠创伤性脑水肿以及水通道蛋白-4表达的影响

摘    要:目的观察甘露醇与尼莫地平联合应用对大鼠挫裂伤脑组织水通道蛋白-4(AQP-4)表达的影响。方法成年Wistar大鼠54只,随机分为挫裂伤组、甘露醇治疗组、甘露醇与尼莫地平联合治疗组。采用文献报告的方法制作大鼠左顶叶皮层局限性脑挫裂伤模型。各组分别于脑挫裂伤后24、48、72h取挫伤区脑组织,检测其含水量以及AQP-4的表达。结果随着伤后时间的增加;各组脑组织含水量增加,AQP-4mRNA和AQP-4蛋白的表达均逐渐增高(P〈0.0.5);与挫裂伤组相比较,甘露醇治疗组伤后各时间点脑组织含水量、AQP-4mRNA和AQP-4蛋白的表达均明显增加(P〈0.05);甘露醇与尼莫地平联合治疗组伤后各时间点与前两组相比,脑组织含水量及AQP-4mRNA和AQP-4蛋白的表达均明显降低(P〈0.05)。结论脑挫裂伤后,AQP-4mRNA和AQP-4蛋白的表达增加;脯组织含水增加,甘露醇联合应用尼莫地平后,能够下调AQP-4mRNA和AQP-4蛋白的表达,有效缓解创伤性脑水肿。

Effects of Mannitol and Nimodipine on Brain Edema and Aquaporin-4 Expression in Rats with Traumatic Brain Injury

Abstract:Objective To observe the effect of mannitol and nimodipine on traumatic brain edema and aquaporin-4 (AQP-4) expression. Methods Fifty-four adult Wistar rats, who suffered from traumatic brain injury(TBI) of the left parietal lobe, were randomly divided into three groups, i.e. blank (no treatment) group (group A), treatment group with mannitol (group B), and treatment group with mannitol and nimodipine (group C). The water content and expression of AQP-4 were determined in the confused brain tissues 24, 48 and 72 hours after TB1 in all the rats. Results The brain water content, and expressions of AQP--4 mRNA and AQP-4 protein gradually increased in all the groups with the lapse of time after TBI (P<0.05). The brain water content, and expressions of AQP-4 mRNA and AQP-4 protein were significantly higher in group B than those in group A after TBI (P<0.05). The brain water content, and expressions of AQP-4 mRNA and AQP-4 protein were significantly lower in group C than those in group A and B after TBI (P<0.05). Conclusions The expressions of AQP-4 mRNA and AQP-4 protein, and the water content increase in the contused brain tissues after TBI. Administration of mannitol combined with nimodipine can inhibit the up-regulation of the expression of AQP-4 mRNA and AQP-4 protein, and can effectively relieve the brain edema in the rats with TBI.
