2-(4-苯氧基苯基)乳酸乙酯染毒与子鼠学习记忆的关系研究

摘    要:目的了解2-(4-苯氧基苯基)乳酸乙酯染毒对子代大鼠学习记忆功能的影响。方法两代繁殖试验大鼠,混饲法染毒,染毒浓度雌性大鼠为0、19.80、80.56和339.49 mg/kg,雄性大鼠为0、18.41、73.74和306.61 mg/kg,子鼠断乳后进行学习记忆功能测试。结果在避暗实验中,F1代子鼠各剂量组比较,错误次数(1.5、0.5、1.0、2.5)和潜伏期(42.5、48.5、26.5、26.0 s),差异均无统计学意义(P>0.05);F2代子鼠各剂量组比较,错误次数(1.5、1.0、1.0、0.5)和潜伏期(13.0、13.5、45.0、20.0 s),差异均无统计学意义(P>0.05)。在穿梭箱实验中,F1代子鼠各组比较,电击次数(8.0、7.0、6.0、7.0)、主动逃避时间(3.0、3.0、4.0、3.5 s),差异均无统计学意义(P>0.05);各组电击时间(15.0、11.0、9.0、13.0 s)两两比较结果显示,尚不能认为各组电击时间差异有统计学意义(P均>0.05)。F2代子鼠各组比较,电击次数(9.0、8.0、8.0、8.0)、电击时间(19.0、18.0、14.0、15.0 s)、主动逃避时间(2.0、2.0、2.0、2.0 s),差异均无统计学意义(P>0.05)。结论在本实验的染毒时间和染毒剂量范围条件下,2-(4-苯氧基苯基)乳酸乙酯对大鼠子代幼鼠学习记忆功能无明显影响。

A Research of Effects of Ethyl 2-(4-phenoxyphenyl) Lactate Exposure on Learning and Memory in Offspring Rats

Abstract:Objective To observe the effects of ethyl 2 - (4 - phenoxyphenyl) lactate exposure on learning and memory of rat offspring. Methods Parent and first offspring (F1) rats were fed with ethyl 2 - (4 - pheuoxyphenyl) lactate at the doses of O, 19. 80, 80. 56 and 339. 49 mg/kg for females and O, 18. 41, 73.74 and 306. 61 mg/kg for males. Functional tests of learning and memory, including step - through test and shuttle - box test, were carried out in the offspring after weaning. Results In passive avoidance test, the F1 rats were compared in each dose, and wrong times ( 1.5,0. 5, 1.0, 2. 5) and latent period (42.5, 48.5, 26.5,26. 0 s) did not differ significantly ( P〉O. 05) ; then offspring rats of F2 were compared in each dose, and wrong times (1.5, 1.0, 1.0, 1. 5) and latent period ( 13.0, 13.5, 45.0, 20. 0 s) did not differ significantly ( P〉 0. 05 ), too. Offspring rats of F1 were compared in each dose in shuttle box experiment, then the number of electric shock (8.0, 7.0, 6. 0, 7.0) and the time of active escaping (3.0, 3.0, 4. O, 3.5) did not differ significantly (P〉0.05), and the time of dectric shock (15.0, 11.0, 9.0, 13.0 s) in pairwise comparison also were not differed significantly ( P〉0. 05). Offspring rats of F2 were compared in each dose in shuttle box experiment, then the number of electric shock (9. 0, 8.0, 8. O, 8.0) , the time of active escaping (2. 0, 2. 0, 2. 0, 2. 0 s) and the time of electric shock (19. 0, 18.0, 14. O, 15.0 s) did not differ significantly ( P〉 0. 05 ). Conclusion Learning and memory function were not damaged in the F1 and F2 rats at the dosages of the test chemical.
