肺癌细胞钙黏蛋白E及连环蛋白的差异表达

摘    要:目的测定不同转移能力肺癌细胞上皮型钙黏蛋白(E-cad),α,β-连环蛋白(α-catenin、β-catenin)的差异表达,探讨它们在肺癌侵袭、转移机制中的作用。方法将高侵袭的肺腺癌细胞(A549)、高转移的肺癌细胞(95-D)、低转移的肺腺癌细胞(SPC-A-1)作常规细胞培养,采用Westen-blot免疫印迹法和逆转录-聚合酶链反应分别从蛋白质和mRNA水平上检测上皮型E-cad及α、β-catenin的表达情况。结果A549与95-D细胞株的E-cad及α、β-catenin表达量较低,95-D表达最弱,二者之间差异无统计学意义(P〉0.05);SPC—A-1细胞株除E-cad弱表达外,α、β-catenin与95-D和A549比较呈明显强表达,差异有统计学意义(P〈0.05)。结论高转移能力的肺癌细胞E-cad和α、β-catenin表达低,低转移的肺癌细胞E-cad和α、β-catenin表达高,表明E-cad及cateinis在肺癌细胞侵袭和转移中起重要作用。

Abstract:Objective To observe the differential expression levels of E-cadherin and α,β-catenins in three lung cancer cell lines with differentially metastatic abilities,in order to study their mechanism of adhesion and metastasis in lung cancer.Methods The three lung cancer cell lines:A549(highly invasive human lung adenocarcinoma cell),95-D(highly metastatic human lung carcinoma cell),and SPC-A-1(lowly metastatic human lung adenocarcinoma cell)were cultured conventionally in vitro.The protein and mRNA expression level of E-cadherin and catenins were observed by western blot analysis and RT-PCR respectively1].Results The total protein and mRNA expression levels of E-cadherin and catenin in A549 and 95-D cells were lower,while the lowest was in 95-D.But there were no significant difference (P>0.05).In SPC-A-1 cell line,the expression level of E-cadherin was low.But the expression levels of catenins were significantly higher than those of the other groups(P<0.05).Conclusion In highly metastatic cells 95-D and A549,the expression levels of E-cadherin and catenins were lower,which lead to invasion and metastasis .In low metastatic SPC-A-1,the expression level was high.The study indicated that E-cadherin and catenins may play an important role in the mechanisms of invasion and metastasis of lung cancer.
