Nav1.1抗体对豚鼠心室肌细胞钠电流的影响

摘    要:目的探讨Nav1.1抗体对豚鼠心室肌细胞钠电流(INa)的影响。方法采用全细胞膜片钳技术,观察急性分离的豚鼠心室肌细胞在应用Nav1.1抗体后心肌INa的变化。结果在Nav1.1抗体作用下,钠离子通道电流密度呈浓度依赖性减小。用药前与三个浓度抗体组(1∶100,1∶50,1∶25)的电流密度分别为34.22±6.22,32.13±3.45,25.49±7.4,19.23±1.69pA/pF。其中1∶25Nav1.1抗体明显降低钠电流密度(P<0.05),并且,1∶25Nav1.1抗体使INa电流-电压曲线明显向正电位方向移动,但是其稳态失活曲线和恢复曲线没有显著性改变。结论Nav1.1可能对INa有影响。

Abstract:Objective To study the effect of antibody of Nav1.1 on sodium current in guinea pig ventricular cells.Methods Detected the sodium current before and after applying antibody of Nav1.1(1∶100,1∶50,1∶25)to the isolated guinea pig ventricular cells with whole cell patch clamp recording.Results Antibody of Nav1.1 decreased the voltage activated Na+ currents in a concentration dependent manner.The density of Na+ current of control was 34.22 ± 6.22pA/pF,while those of test were 32.13 ± 3.45pA/pF,25.49 ± 7.4pA/pF,and 19.23 ± 1.69pA/pF respectively.Sodium current was inhibited by 1∶25 anti-Nav1.1 significantly.1∶25 anti-Nav1.1 shifted I-V relationship curve to more positive potentials.However,anti-Nav1.1 didn't change the steady-state recovery and inactivation of Na+ currents.Conclusions Sodium channel subtypes Nav1.1 contributes to the sodium current in ventricular cells.
