黄芪多糖对大鼠脑缺血再灌注后迟发性神经元死亡影响的实验研究

摘    要:目的:观察黄芪多糖(APES)对大鼠脑缺血再灌注后海马CA1区迟发性神经元死亡(DND)的形态学影响,并探讨黄芪多糖对脑缺血损伤的保护机制。方法:用重复双侧颈总动脉夹闭方法制作不完全前脑缺血再灌注动物模型。于造模后3天取材,HE染色及原位DNA末端标记法观察模型组和APES治疗组动物脑缺血再灌注后海马神经细胞迟发性神经元死亡的情况。结果:造模后3天,模型组海马锥体细胞出现广泛的细胞坏死,以CA1区和齿状回最明显。TUNEL染色法显示模型组CA1区坏死神经元的邻近部位以及齿状回可见大量特征性阳性颗粒的凋亡细胞,治疗组坏死及凋亡细胞明显少于模型组(P<0.05)。结论:黄芪多糖减少缺血再灌注对神经细胞的损害,对缺血再灌注后神经元迟发性死亡具有保护作用。

Abstract:Objective:To observe the effects of APESon the neuronal cell injury in the hippocampal CAl sector following cerebral ischemia reperfusion in rats and to approachthe protectionmechanismof APES onthe neuronal cell injury.Methods: With replicate cerebral ischemia reperfusion rat model,using HE and TUNEL staining,to observe the DND in the hippocampuswith model group and treatment group.Results: In the model group,appear extensive cell death and apoptosis calls in the hippocampal pyramidal cells on CA1 region and dentate gyrus. The structural damage reducedmarkedlyin the treatmentgroup than model group.Conclusion:APES saporins could distinctly relieve the neurotic reaction and the injured degree of neuron in hippocampus CAI.
