酒石酸锑钾对人结肠癌裸鼠移植瘤生长的抑制作用

摘    要:目的:观察酒石酸锑钾(PAT)对人结肠癌裸鼠移植瘤的抑瘤及凋亡诱导作用。方法:60只裸鼠皮下注射结肠癌SW480细胞建立移植瘤模型,随机分为4组,每组15只,分别腹腔内注射生理盐水、5-Fu及不同剂量的PAT,给药15 d,末次给药24 h后处死裸鼠,计算抑瘤率并观察肿瘤细胞的凋亡。用末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL法)检测肿瘤细胞凋亡。结果:PAT对人结肠癌裸鼠移植瘤生长有显著性抑制作用,肿瘤细胞凋亡增加;高剂量PAT组、低剂量PAT组和5-Fu组的凋亡指数高于生理盐水组,差异有显著性(P<0.05),高剂量PAT组的凋亡指数高于5-Fu组,差异有显著性意义(P<0.05)。结论:酒石酸锑钾可通过诱导结肠癌SW480细胞凋亡抑制结肠癌裸鼠移植瘤生长。

Abstract:Objective: To study the potential therapeutic effects of potassium antimonyl tartrate(PAT) on heterologous graft mice models with human colon neoplasm.Methods: To build transplanted animal models,sixty nude mice were inoculated with colon cancer cells SW480.Nude mice were then randomly divided into four groups and administrated with saline,5-Fu and different dose of PAT respectively for 15 d.After final-administration for 24 h,mice were killed and the tumor inhibition rate(TIR) was calculated.Cell apoptosis in situ was examined by a TUNEL assay in the samples of carcinoma.Results: PAT inhibited the growth of transplanted tumor significantly and the TIR was increased.The AI of 20 mg/(kg·d) PAT group and 40 mg/(kg·d) PAT group was much more than that of normal saline respectively.Conclusion: PAT can significantly inhibits the transplanted tumor growth in nude mice bearing colon carcinoma,mainly through inducing cell apoptosis.
