奥沙利铂与常用化疗药物对胃癌细胞株相互作用的研究

目的　利用胃癌细胞株探讨奥沙利铂与常用化疗药物的相互作用。方法　测定奥沙利铂与 4种常用化疗药的单药和联合用药对胃癌细胞株的抑制率 ,将测得的数据输入电脑 ,用CalcuSyn(Biosoft)软件处理 ,推算出有关数据 :斜率(m )、截距 ( y int)、IC50 、联合指数 (CI)和剂量减少指数 (DRI)。结果　胃癌细胞株单药化疗敏感性顺序 :OXA >MMC >5 Fu >FUDR >CDDP ;联合用药的敏感性顺序 :CDDP +OXA >FUDR +OXA >5 Fu +OXA >MMC +OXA。联合用药协同作用的大小依次为 :FUDR +OXA >5 Fu +OXA >MMC +OXA。根据这 4组两种药物联合用药的CI和DRI综合分析 ,以FUDR +OXA和 5 Fu +OXA组较佳 ;此外 ,三种药物联用的CI和DRI也十分理想。结论　OXA与常用化疗药物联合 ,对胃癌细胞株有良好的抑制率

Study on the Interaction among Oxaliptin and Common Chemotherapy Drugs in Gastric Cancer Cell Line

Objective To evaluate the interaction among oxaliptin(OXA) and common chemotherapy drugs using gastric cancer cell line.Method MTT assay was employed to determine the cytotoxicity(inhibition rate) of OXA,5-FU,FUDR,MMC and CDDP in gastric cancer cell line(SGC7901).The results from all experiments were analysed with CalcuSyn(Biosoft) software.The associated parameters:m,y-int,IC 50,CI and DRI were concluded.Results Chemosensitivity sequence for single drug was OXA>MMC>5-FU>FUDR>CDDP,chemosensitivity sequence for combined drug was CDDP+OXA>FUDR+OXA>5-FU+OXA>MMC+OXA.Synergistic effect sequence for combined drug was FUDR+OXA>5-FU+OXA>MMC+OXA.FUDR+OXA and 5FU+OXA were more effective in two drugs combined chemotherapy according to the analysis of CI and DRI.In addition,the combined chemotherapy of three drugs was also effective.Conclusions The combined chemotherapy of OXA and common chemotherapy drugs is effective on gastric cancer cell line in vitro.
