利凡诺的遗传毒性研究

摘    要:采用Ames试验,小鼠骨髓多染红细胞(PCE)微核试验,中国仓鼠肺细胞(CHL)染色体畸变试验,人外周血淋巴细胞染色体畸变试验和姐妹染色单体互换(SCE)试验对利凡诺进行检测。结果利凡诺在TA97、TA100和TA102菌株±S9mix时剂量1~100μg/皿阴性,TA98菌株-S9阴性,+S9阳性,显示出诱变性。各剂量组微核细胞率0.9‰~1.2‰,与溶剂对照1.0%比较,无显著差异(P>0.05),3个剂量组CHL细胞染色体畸变率在正常范围(<5%)。不同时相人外周血淋巴细胞染色畸变率和SCE频率分别与对照比较,无显著差异(P>0.05)。研究结果表明利凡诺无诱发小鼠骨髓PCE微核、CHL细胞染色体畸变、人外周血淋巴细胞染色体畸变和SCE频率的增加作用。仅发现利凡诺对TA98菌株(+S9mix)有诱发回复突变菌落数增加作用,是一种细菌致突变剂,但对人体不产生潜在遣传危害。

Abstract:bjectives:To study the genetoxicity of rivanol in order to evaluate its safety. Settings:Genetic toxicology laboratory,Chongqing Family Planning Scientific Research Institute. Methods:The strains TA97,TA98,TA100 and TAl02 with or without S9mix in the Ames test,the micronucleus assay of polychromatic erythrocytes in mice,the chromosome aberration assay in Chi-nese hamster lung cells(CHL),the chromosome aberration assay and sister chromatid exchange(SCE)in peripheral lymphocytes of human Were measured. Results:The results showed that rivanol in 5 doses of 1~100μg/plate did not induce positive muta-tion of strains TA97,TA100 and TA102 with or without S9mix in the Ames test.but it increased thenumber of revertants of strains TA98 with S9mix,The rates of micronucleus cells of each rivanol doseswere 0.9‰~1.2‰,and no significant difference was found as compared with l.0‰ of solvents group.The rates of CHL chromosome aberrations of rivanol in doses of 7.5,15 and 30μg/ml were normal(P<5%).The rates of chromosome aberration cells in human peripheral lymphocytes in all rivanol groupswere between 2.1%~2.2%,there was no significant difference as compared with the control group(P>0.05).The means of SCEs of two rivanol groups were between 5.71~5.79 the difference was not sig-nificant(P>0.05).when compared with the control group.Conclusion:The study results showed that rivanol was not mutagenic in micronucleus assay,chro-mosome aberration and SCE assay,but mutagenic for strain TA98.Rivanol has no potential genetic harmto human and is safe when administered in clinic.
