高迁移率族蛋白1在大鼠动脉粥样硬化斑块中表达的研究

摘    要:目的 观察动脉粥样硬化大鼠高迁移率族蛋白1(HMGB-1)的蛋白及基因表达变化,以揭示与炎症相关的动脉粥样硬化发病机制.方法 免疫组织化学染色法观察大鼠主动脉血管动脉粥样硬化斑块内HMGB-1的蛋白表达情况,RT-PCR法检测大鼠主动脉血管HMGB-1的基因表达变化.结果 正常血管壁内存在HMGB-1表达,位于血管内皮细胞、平滑肌细胞和成纤维细胞的细胞核内 动脉粥样硬化斑块内HMGB-1分布不均匀,炎症细胞浸润区和坏死区周边基质内表达明显增高,炎症细胞浸润区内以单核-巨噬细胞胞核、胞浆和间质内表达为主,后者成条带状分布于坏死区边缘.动脉粥样硬化斑块内HMGB-1水平均明显升高,与正常血管壁比较差异有显著性(P〈0.01).结论 动脉粥样硬化大鼠主动脉血管HMGB-1的蛋白及基因表达均明显增强.

Abstract:Objective To observe the protein and gene expression of high mobility group box-1(HMGB-1)in atherosclerotic rats,and to discover the mechanisms of atherosclerosis related to inflammation.Methods 20 healthy male rats were randomly divided into normal control group(n=10) and atherosclerosis group(n=10).Animals in atherosclerosis group were fed with high fat diet for 10 w and given intragastrically vitamin D3 at 400 000 U/kg once a week for successive three times,while those in normal control group was fed with normal diet.After 10 w of feed,the blood sample was collected from the abdominal aorta of each rat for the measurement of the expression changes of HMGB-1 gene by real time PCR.Meanwhile,the expression of HMGB-1 protein in aortal atherosclerotic plaques was detected by immunohistochemistry.Results In normal vessels,HMGB-1 protein was mostly expressed in the nuclei of vascular endothelial cells,smooth muscle cells and fibroblasts.Nevertheless,it was unevenly located in the atherosclerotic plaques with significantly increasing expression in inflammatory cells-infiltrated areas and the matrix around the necrotic areas.In inflammatory cells-infiltrated areas,HMGB-1 protein was expressed mainly in the nuclei and cytoplasm of monocyte- macrophages as well as stroma while in the necrotic areas,the stripped expression of HMGB-1 protein was mainly distributed in the edge.HMGB-1 protein level was higher in atherosclerotic plaques than that in normal vessels (P〈0.01).Conclusion Both gene and protein expression of HMGB-1 in the aortal vessels of atherosclerotic rats appear to increase obviously.
