S100A4与卵巢癌细胞顺铂耐药相关性的研究

目的:探讨钙结合蛋白S100A4与卵巢癌细胞顺铂耐药的关系。方法:MTT法测定顺铂对卵巢癌顺铂敏感细胞株A2780和耐药株CP70的半效抑制浓度(IC50);分别应用免疫细胞化学、Western blot和实时荧光定量PCR检测A2780和CP70细胞株中S100A4的差异表达,观察顺铂处理CP70细胞后S100A4在蛋白水平及基因水平的动态变化。结果:顺铂对A2780和CP70的IC50分别为20.88μmol/ml和57.10μmol/ml。S100A4蛋白在二者中均以胞浆表达为主;CP70中S100A4 mRNA水平和蛋白水平的表达均显著高于A2780;经顺铂处理CP70后,S100A4的表达呈剂量-时间依赖性。结论:S100A4的高表达与卵巢癌顺铂化疗耐药有关。

Relationship between S100A4 expression and platinum-based chemotherapy resistance in ovarian carcinoma

Objective:To investigate the correlation between S100 calcium-binding protein A4(S100A4)and platinum-based chemotherapy resistance in ovarian carcinoma.Methods:MTT assay was performed to determine the 50% inhibitory concentration(IC50)to cisplatin in the cisplatin sensitive human ovarian cancer cell line A2780 and cisplatin resistance cell line CP70.The expressions of S100A4 in A2780 and CP70 cells were identified by immunocytochemical staining,Western blot and real-time PCR,respectively.The changes of S100A4 expressions in CP70 cells pretreated with cisplatin were detected by real-time PCR and Western blot.Results:The IC50 was 20.88μmol/ml in A2780 cells.The IC50 was 57.10μmol/ml in CP70 cells.S100A4 was mainly located in the cytoplasm of A2780 and CP70.real-time PCR and Western blot showed that the expressions of mRNA and protein of S100A4 were significantly higher in CP70 cells than in A2780 cells.In CP70 cells cisplatin treatment reduced the expressions of mRNA and protein of S100A4 in a dose-and time-dependent manner.Conclusion:The expression level of S100A4 may be correlated with cisplatin resistance in ovarian carcinoma.
