重度子痫前期患者血清差异蛋白质组学研究

摘    要:目的:利用蛋白质组学技术寻找敏感性高、特异性好的血清标志物作为重度子痫前期的诊断依据。方法:采用双向凝胶电泳与质谱联用的方法筛选子痫前期重度患者和健康孕妇血清中的差异蛋白质。2-DE分离两组血清蛋白质,图像分析软件识别差异蛋白质点,电喷雾串联质谱(ESI-Q-TOF MS/MS)鉴定差异蛋白质,Western blot检测差异蛋白质—P物质在两组血清中的表达情况。结果:采用双向凝胶电泳与质谱识别了8个差异蛋白质点,鉴定出3种差异蛋白:凝血酶、P物质及白蛋白。Western blot结果显示,子痫前期重度患者血清中P物质的表达水平明显低于健康孕妇。结论:凝血酶、P物质及白蛋白可能参与了重度子痫前期的发生、发展过程,检测它们的水平对子痫前期重度的诊断可能具有一定的参考价值。

Abstract:Objective:To investigate the high sensitive and specific serum markers for diagnosis the onset of severe pre-eclampsia by proteineomacs technique.Methods:Two-dimensional gel electrophoresis and mass spectrometry were used to screen the difference of serum proteins between severe pre-eclampsia patients and healthy control group.The serum proteins of the two groups were separated by 2-DE method.Using the image analysis software to analysis the different protein spots.Different protein was identified by the electro-spray ionization tandem mass spectrometry(ESI-Q-TOF MS/MS).The expression of substance P protein was analyzed by Western blot.Result:Eight different protein spots were identified by 2-DE and mass spectrometry.Three different proteins which thrombin,substance P and albumin protein were discriminated.The expression level of substance P in serum of patients with severe pre-eclampsia was significantly lower than that in the control group.Conclusions:Thrombin,substance P and albumin protein may be invovled in the the development of onset of severe pre-eclampsia.It can be a useful marker to diagnose onset of severe preeclampsia.
