结核病患者氧氟沙星的群体药代动力学

摘    要:背景:美国两所结核病医院。 目的:测定口服多种剂量氧氟沙星后的群体药代动力学(PK)。 设计:共73例结核病患者参加本研究,其中一部分患者接受多种剂量的氧氟沙星。根据药敏结果,同时服用其它药物。收集10小时以上的血清样本经高压液相色谱仪测定,采用群体方法分析浓度-时间数据。 结果:氧氟沙星浓度随增加的口服剂量呈线形上升。29%患者至少有一次吸收延缓。氧氟沙星清除随肾功能下降和年龄增加而降低。患者对较高剂量耐受良好,且可最大限度提高血药峰值浓度与最低抑菌浓度之比值(Cmax:MIC)。 结论:氧氟沙星群体药代动力学参数与以往报告结果相似。较高剂量可具有良好的药代动力学,有利于结核病的治疗。
