载氧预适应对小鼠缺氧耐受能力的影响

摘    要:目的观察载氧预适应对小鼠急性缺氧耐受能力的影响。方法观察腹腔注射氧气后小鼠密闭缺氧耐受时间、亚硝酸钠中毒存活时间、断头后张口动作持续时间、氰化钠中毒存活时间、丙二酸中毒存活时间及碘乙酸中毒存活时间,并与腹腔注射空气小鼠相比较。结果腹腔注射氧气3 d组(1 ml/只,1次/d)和3 h组(1 ml/只)密闭缺氧耐受时间分别显著高于腹腔注射空气3 d和3 h组(P〈0.05);腹腔注射氧气3 h组小鼠断头后张口动作持续时间和氰化钠中毒存活时间显著高于腹腔注射空气组(P〈0.05),亚硝酸钠中毒、丙二酸中毒和碘乙酸中毒存活时间与腹腔注射空气组没有显著性差异(P〉0.05)。结论载氧预适应可显著提高小鼠缺氧耐受能力,其机制可能与提高血氧饱和度改善机体缺氧状况有关,同时也提示三羧酸循环和糖酵解途径等可能参与了载氧预适应的抗缺氧作用。

Abstract:Objective To observe the effects of oxygen-loaded preconditioning on hypoxic tolerance in mice.Methods 41 mice were randomly divided into 3 h-oxygen-loaded group and 3 d-oxygen-loaded group,3 h-air-loaded group and 3 d-air-loaded group.In these four groups,1 ml of oxygen or air per mouse was injected into abdominal cavity only once or once daily for 3 d.At 3 h after injection or the last injection,they submitted to a series of tests.Hypoxic tolerance time in a sealed bottle,survival time after administration of nitrite,sodium cyanide,malonic acid and iodoacetic acid as well as gape duration after decapitation were recorded and compared among the four groups.Results Hypoxic tolerance time in a sealed bottle was significantly longer in 3 d-and 3 h-oxygen-loaded group than that in 3 d-and 3 h-air-loaded group,respectively(P0.05).Gape duration after decapitation and survival time of sodium cyanide poisoning were significantly longer in 3 h-oxygen-loaded group than that in 3 h air-loaded group(P0.05),but no statistical difference was found in survival time after administration of nitrite,malonic acid and iodoacetic acid between these two groups(P0.05).Conclusion Oxygen-loaded preconditioning can promote hypoxic tolerance of mice probably due to the elevation of blood oxygen saturation,and regulation of tricaboxylic acid cycle and glycolytic pathways.
