摘    要:目的 应用不同频率电刺激大鼠束旁核 (PF) ,观察对黑质网状部 (SNR)神经元自发放电频率的影响 ,探讨PF对SNR神经元活动的调节作用。方法 采用单管玻璃微电极细胞外记录方法 ,观察SNR神经元自发放电频率在不同频率 (强度 0 4mA ,波宽 0 1ms,时程 5s ,频率 1Hz、 10Hz、 2 0Hz、 5 0Hz、 80Hz、 10 0Hz、 130Hz、 15 0Hz、 180Hz、 2 0 0Hz、 2 5 0Hz、 30 0Hz和 5 0 0Hz)电刺激PF时的变化。结果 高频电刺激抑制93 6 7%SNR神经元的自发放电频率 ,抑制时程具有频率依赖性 ,有效电刺激频率需大于 5 0Hz,且随外加刺激频率增加 ,抑制时程延长 ,二者呈正相关。刺激频率超过 2 0 0Hz后 ,抑制时程保持恒定 ,不再随外加刺激频率而发生显著变化。结论 高频刺激PF ,可以降低基底节输出核团SNR的神经元活动 ,本研究提示高频刺激PF对帕金森氏病的运动症状有治疗作用。

Abstract:Objective To research the effect of the electrical stimulation of the parafascicular nucleus (PF) on the neural activities of the subtantia nigra (SNR) in rats. Methods Extracellular recording were used to observe the changes of the firing rates and patterns of the SNR to the electrical stimulation (width: 0.1ms, intensity: 0.4mA, duration:5s, frequency:1,10,20,50,80,100,130,150,180,200,250,300 and 500Hz) of the PF . Results Electric stimulation of the PF inhibited the majority firing rates of the neuron in SNR (93.67%). Moreover, the inhibitory duration was in a frequency-dependent manner. Under 50Hz, there is no effect and when the stimulative frequency was more than 50Hz, the inhibition duration changed with the stimulation frequency. Conclusions High-frequency stimulation of the PF induced an inhibitory effect on the spontaneous activity of the SNR in normal rats. This result suggest that HFS on PF should be an effective surgical procedure for the treatment of Parkinson's disease.
